目的:探讨兔蛛网膜下腔出血脑血管痉挛前后基底动脉中PDEⅤ的表达变化。方法:制作脑血管痉挛动物模型,空白对照组、实验对照组与SAH组均于制模后第3天行TCD与选择性椎-基底动脉造影,检测兔脑血管痉挛的发生及变化,检测完后,取其基底动脉,制成病理切片备用。通过免疫组化检测兔基底动脉中PDEⅤ的表达。结果:各配伍组中TCD测得的基底动脉血流速度、DSA测得的基底动脉管径值及免疫组化阳性表达产物的SUM值不等或不全相等(P<0.05),经Student-Newman-Keuls检验对各配伍组数据进行两两比较,SAH组与空白对照组、SAH组与实验对照组TCD测得的基底动脉血流速度、DSA测得的基底动脉管径值及免疫组化阳性表达产物的SUM值不等(P<0.05),空白对照组与实验对照组TCD测得的基底动脉血流速度、DSA测得的基底动脉管径值及免疫组化阳性表达产物的SUM值差异无统计学意义(P>0.05)。结论:兔蛛网膜下腔出血脑血管痉挛后PDEⅤ在其基底动脉中明显表达,推测PDEⅤ表达可能与脑血管痉挛发生有关。

목적:탐토토주망막하강출혈뇌혈관경련전후기저동맥중PDEⅤ적표체변화。방법:제작뇌혈관경련동물모형,공백대조조、실험대조조여SAH조균우제모후제3천행TCD여선택성추-기저동맥조영,검측토뇌혈관경련적발생급변화,검측완후,취기기저동맥,제성병리절편비용。통과면역조화검측토기저동맥중PDEⅤ적표체。결과:각배오조중TCD측득적기저동맥혈류속도、DSA측득적기저동맥관경치급면역조화양성표체산물적SUM치불등혹불전상등(P<0.05),경Student-Newman-Keuls검험대각배오조수거진행량량비교,SAH조여공백대조조、SAH조여실험대조조TCD측득적기저동맥혈류속도、DSA측득적기저동맥관경치급면역조화양성표체산물적SUM치불등(P<0.05),공백대조조여실험대조조TCD측득적기저동맥혈류속도、DSA측득적기저동맥관경치급면역조화양성표체산물적SUM치차이무통계학의의(P>0.05)。결론:토주망막하강출혈뇌혈관경련후PDEⅤ재기기저동맥중명현표체,추측PDEⅤ표체가능여뇌혈관경련발생유관。
Objective:To study the basilar artery expression changes of PDEⅤbefore and after cerebral vasospasm in subarachnoid hemorrhage rabbits. Method:Made the vasospasm rabbits model.In the all groups,the DSA and TCD were made to assess the cerebral vasospasm on the 3rd day. After that,sacrifice the animal models,and then take the basal arteries as the specimen. The specimen were made paraffin-embedded sections for Immunohistochemical examination. Result:The velocity of basal arteries by TCD﹑the arteries diameters by DSA and the SUM by IHC in all groups were not equal or not completely equal(P<0.05). And Significantstatistics differences were found in multiple comparison(P<0.05),between SAH and blank contrast group,SAH and experimental contrast group by Student-Newman-Keuls test.But no differences were found between blank and experimental contrast group(P>0.05). Conclusion:The PDE Ⅴ express obviously in basilar artery of subarachnoid hemorrhage rabbits. So the PDEⅤand the CVS may be closelyy related.
