目的:探讨栓塞猪冠状动脉前降支制备急性心肌梗死动物模型的可行性及其技术分析。方法健康成年雄性滇南小耳猪12头,麻醉后行股动脉穿刺,插管至冠状动脉行左、右冠状动脉造影术,采用自制可脱球囊永久植入冠状动脉前降支(left anteri-or descending branch,LAD)中远1/3段,术中行心电动态监护,并分别于栓塞术前、术后6 h、24 h、72 h及1周行心肌酶学、心肌坏死标记物检测,全部动物于1周后处死,行TTC染色及病理切片分析以评价模型建立是否成功。结果11头猪饲养至实验终止,经心电图、相关实验室检测及病理染色证实模型制备成功,且显示梗死部位均集中在心尖段、左室前壁心尖段,部分模型累及室间隔心尖段。其中1头猪于术中发生室颤,抢救无效死亡。结论运用自制可脱球囊栓塞 LAD可成功建立猪急性心肌梗死模型,该方法模型制备安全性较高,梗塞部位集中,各项观察指标表现明确,病理结果肯定,是急性心肌梗死实验研究的理想动物模型。

목적:탐토전새저관상동맥전강지제비급성심기경사동물모형적가행성급기기술분석。방법건강성년웅성전남소이저12두,마취후행고동맥천자,삽관지관상동맥행좌、우관상동맥조영술,채용자제가탈구낭영구식입관상동맥전강지(left anteri-or descending branch,LAD)중원1/3단,술중행심전동태감호,병분별우전새술전、술후6 h、24 h、72 h급1주행심기매학、심기배사표기물검측,전부동물우1주후처사,행TTC염색급병리절편분석이평개모형건립시부성공。결과11두저사양지실험종지,경심전도、상관실험실검측급병리염색증실모형제비성공,차현시경사부위균집중재심첨단、좌실전벽심첨단,부분모형루급실간격심첨단。기중1두저우술중발생실전,창구무효사망。결론운용자제가탈구낭전새 LAD가성공건립저급성심기경사모형,해방법모형제비안전성교고,경새부위집중,각항관찰지표표현명학,병리결과긍정,시급성심기경사실험연구적이상동물모형。
Objective Aimed to probe the feasibility and technology about the preparation of swine acute myocardial infarction model by embolizing the left anterior descending branch of coronary artery.Methods 1 2 healthy and mature male Diannan tiny-ear pigs were selected.Implanted self-made sacculus to their LAD permanently after femoral artery puncture and coronary angiography. In the course of operation,electrocardiograph monitoring was applied.Marker detections were performed pre-operation,post-opera-tion 6h,24h,72h,and 1w respectively.All the selected pigs were killed after a week.Then the TTC staining and pathological sec-tion were applied to analyze and access the possibilities of building model.Results 1 1 pigs kept alive till the end of the experiments. It was proved that the preparation of model was successful through electrocardiograph,lab inspection and pathological staining.The result unveiled that the embolism positions were at cardiac apex and apica sections of left ventricular in all the tested pigs,interven-tricular septum was involved in some models.One pig dead owing to ventricular fibrillation during the operation.Conclusion It is possible to build swine acute myocardial infarction model using self-made sacculus.High safety,concentrated infarction areas,clear indicators and certain pathological results were acquired.It is a feasible animal model for studying acute myocardial infarction.
