目的探讨HIF-1α、bFGF、Survivin在慢性高原病(CMS)发展过程中的意义及其对血管生成的影响。方法应用免疫组织化学 SP法检测 HIF-1α、bFGF及Survivin在30例CMS患者(CMS组)及20例长期居住西宁地区健康人(对照组)骨髓组织中的表达,并计数CD34标记骨髓组织的微血管密度(MVD)结果 HIF-1α、bFGF及Survivin 在CMS组阳性表达率均显著高于对照组(P值均<0.01);MVD在CMS组也明显高于对照组(P<0.01)。HIF-1α、bFGF、Survivin在CMS组的阳性表达率、MVD及Hb相互间均呈显著正相关(均为P<0.01)。结论 HIF-1α、bFGF和Survivin 在CMS骨髓组织中表达升高, MVD增加,提示 Survivin可能通过抑制细胞凋亡,与HIF-1α及bFGF协同促进血管生成和红细胞增殖,参与CMS的发生发展。

Objective To Investigate the roles of HIF-1α and bFGF and Survivin in development of chronic mountain sickness (CMS) and their ef ects on angiogenesis. Methods The samples of bone marrow used in this study were obtained from 30 patients with CMS (CMS group)and 20 healthy adults (control group). The expressions of HIF-1α and bFGF and Survivin in bone marrows were detected using the SP immunohistochemistry technique and the MVD in bone marrow was measured by counting the vascular endothelial cel s labeled by anti-CD34 monoclonal antibody. The results were analyzed comparatively. Results The positive rates of HIF-1α and bFGF and Survivin in CMS group were significant higher than that in control group (P<0.01). MVD in CMS groupwas significantly higher than that in control group (P<0.01) .In the CMS group,there were positive correlations in the positive rates of HIF-1αand bFGF and Survivin, MVD and Hb each other (al P<0.01). Conclusions The results of this study showed that there were over expressions of HIF-1αand bFGF and Survivin, and the MVD increased in bone marrow of CMS. It was suggested that the HIF-1αand bFGF and Survivin may play a crucial role in generation and progression of CMS through contributing to the angiogenesis and the erythrocyte proliferation in CMS .
