目的:观察人参皂苷 Rg1对鼠胚成纤维细胞(MEF)向诱导多潜能干细胞(iPSCs)诱导转化效率的影响。方法:采用经典四因子的逆转录病毒载体感染,在细胞感染后3 d 内在 MEF 培养基里加入人参皂苷Rg1(0、0.3、1、3、10滋g/mL),进行 iPSCs 诱导及鉴定。结果:人参皂苷 Rg1(1滋g/mL)组可明显提高 iPSCs 的诱导效率,诱导效率为(0.0450依0.0019)%,人参皂苷 Rg1(0滋g/mL)组为(0.0100依0.0033)%,所获 iPSCs 经鉴定为阳性。结论:人参皂苷 Rg1可能在提高小鼠 iPSCs 的诱导效率方面发挥一定作用。

목적:관찰인삼조감 Rg1대서배성섬유세포(MEF)향유도다잠능간세포(iPSCs)유도전화효솔적영향。방법:채용경전사인자적역전록병독재체감염,재세포감염후3 d 내재 MEF 배양기리가입인삼조감Rg1(0、0.3、1、3、10자g/mL),진행 iPSCs 유도급감정。결과:인삼조감 Rg1(1자g/mL)조가명현제고 iPSCs 적유도효솔,유도효솔위(0.0450의0.0019)%,인삼조감 Rg1(0자g/mL)조위(0.0100의0.0033)%,소획 iPSCs 경감정위양성。결론:인삼조감 Rg1가능재제고소서 iPSCs 적유도효솔방면발휘일정작용。
ObjectiveTo observe the effects of Ginsenoside Rg1 on the induce efficiency of mouse induced pluripotent stem cells (iPSCs) from Mouse embryonic fibroblasts (MEF). Methods: The cells were infected with classic retro-viruses vector expressing classic 4 factors, then Ginsenoside Rg1 (0 、0.3、1、3、10 μg/mL) was added to the MEF medium during the first 3 days after infection. Then the clones were identified. Results: Ginsenoside Rg1 (1 μg/mL)can obviously improve the induction efficiency of iPSCs [(0.0450±0.0019)% in the Ginsenoside Rg1group vs (0.0100±0.0033)% in the control group)]. The clones were positive. Conclusion: Ginsenoside Rg1 may facilitate the inducing efficiency of iPS cells.
