目的:观察他莫昔芬(TAM)对氧糖剥夺(OGD)模型中小胶质细胞 BV-2增殖和迁移的影响。方法:将BV-2细胞分为对照组(常规正常氧浓度+高葡萄糖+无胎牛血清培养基)、OGD 组(1%氧浓度+无糖+无血清培养基)、OGD+TAM 组(1%氧浓度+无糖+无血清培养基+TAM),采用划痕试验,通过免疫荧光技术检测小胶质细胞 BV-2在不同氧浓度下的增殖和迁移情况。同时观察 TAM 对小胶质细胞活化的调节作用。结果:与对照组相比,BV-2在 OGD 后细胞迁移加快(<0.05);OGD+TAM 组迁移较 OGD 组下降(<0.05);OGD 组 Ki67阳性率的 BV-2明显高于对照组(<0.05);OGD+TAM 组 Ki67阳性率则明显低于 OGD 组(<0.05)。结论:缺氧可诱导小胶质细胞活化,雌激素受体拮抗剂 TAM 能有效抑制这种活化。

목적:관찰타막석분(TAM)대양당박탈(OGD)모형중소효질세포 BV-2증식화천이적영향。방법:장BV-2세포분위대조조(상규정상양농도+고포도당+무태우혈청배양기)、OGD 조(1%양농도+무당+무혈청배양기)、OGD+TAM 조(1%양농도+무당+무혈청배양기+TAM),채용화흔시험,통과면역형광기술검측소효질세포 BV-2재불동양농도하적증식화천이정황。동시관찰 TAM 대소효질세포활화적조절작용。결과:여대조조상비,BV-2재 OGD 후세포천이가쾌(<0.05);OGD+TAM 조천이교 OGD 조하강(<0.05);OGD 조 Ki67양성솔적 BV-2명현고우대조조(<0.05);OGD+TAM 조 Ki67양성솔칙명현저우 OGD 조(<0.05)。결론:결양가유도소효질세포활화,자격소수체길항제 TAM 능유효억제저충활화。
Objective To observe the effects of tamoxifen (TAM) on the migration and proliferation of BV-2 cells in oxygen-glucose deprivation (OGD) models. Methods: BV-2 cells were divided into control group (normal oxygen concentration+ high glucose+ fetal bovine serum free medium), OGD group (1% oxygen + sugar free+serum-free medium), OGD+TAM group (1% oxygen + sugar free+serum-free medium +TAM). After performing scratch test, migration and proliferation of BV-2 cells were examined by photographs and immunohistochemistry. Results: BV-2 cells in the OGD group shown faster migration than those in the control group ( <0.05). The administration of TAM suppressed the migration of BV-2 cells compared to the OGD group ( <0.05). The percentage of Ki67 positive cells in the OGD group was higher than that in the control group, and the application of Tam reduced the percentage of Ki67 positive cells compared with that in the OGD group ( <0.05). Conclusion:Tamoxifen may play a neuro-protective role in hypoxic condition by suppressing the migration and proliferation of BV-2 cells after OGD.
