目的:研究复方丹参片对阿尔茨海默病(Alzheimer disease,AD)转基因细胞模型微管相关蛋白(Tau蛋白)表达的影响。方法将SD大鼠随机分成复方丹参片0.5、1、2、4倍剂量组、空白对照组及多奈哌齐组,分别灌服相应药液,空白组给予等容积生理盐水,1次/日,连续7 d之后,取各组血清培养AD转基因细胞,于24、48、72 h采用Western Blot法分析AD转基因细胞Tau蛋白表达情况。结果与空白对照组相比,24 h时复方丹参片1、2、4倍剂量组能降低Tau蛋白表达,差异有统计学意义(P<0.01);48 h时复方丹参片2、4倍剂量组差异有统计学意义(P<0.01);72 h时各剂量组间差异均无统计学意义(P>0.05)。结论复方丹参片2倍剂量48 h时能显著减少AD转基因细胞模型Tau蛋白表达。

목적:연구복방단삼편대아이자해묵병(Alzheimer disease,AD)전기인세포모형미관상관단백(Tau단백)표체적영향。방법장SD대서수궤분성복방단삼편0.5、1、2、4배제량조、공백대조조급다내고제조,분별관복상응약액,공백조급여등용적생리염수,1차/일,련속7 d지후,취각조혈청배양AD전기인세포,우24、48、72 h채용Western Blot법분석AD전기인세포Tau단백표체정황。결과여공백대조조상비,24 h시복방단삼편1、2、4배제량조능강저Tau단백표체,차이유통계학의의(P<0.01);48 h시복방단삼편2、4배제량조차이유통계학의의(P<0.01);72 h시각제량조간차이균무통계학의의(P>0.05)。결론복방단삼편2배제량48 h시능현저감소AD전기인세포모형Tau단백표체。
Objective Study on the expression of the Tau that ADtransgenic cell has been cultured by Danshen Tablets (DT). Methods SD rats were randomly divided into 6 groups, including 0.5 dose, 1.0 dose, 2.0 dose, 4.0 dose DT groups, blank control group and donepezil group. Each group rats respectively were given different concentrations of drugs, blank group was given equal normal saline, 1 time a day, 7 days in a row . Then each containing medicine serum of rats were taken to cultivate the AD transgenic cells, and using the m ethods that Western blot to test the levels of Tau protein after cultured 24h, 48h, 72h. Results Compared with the blank control group, At 24h time point, the expression of Tau were declined by 1×, dose of DT , there were significant effects (P<0.01);At 48h, 2×and 4× dose of DT , it still had significant effects (P<0.01); At 72h, there are non-significant effect with all the dose groups of DT (P>0.05). Conclusion 2 × dose of DCT could reduce the level of Tau obviously at 48h on the AD transgenic cell model.
