为了解鲢( Hypophthalmichthys molitrix)、鳙( Aristichthys nobilis)在千岛湖中的食性差异,对千岛湖鲢、鳙肠道内的原核生物16S rDNA和真核生核18S rDNA中的可变区进行PCR-DGGE指纹图谱分析。分别得到53个原核生物的操作分类单元( operational taxonomic units , OTUs)和45个真核生物的OTUs。基于Dice相似系数进行的聚类以及群落的主成分分析( PCA)显示,鲢、鳙肠道内原核生物种群略有重叠,而真核生物种群差异明显。另外,对13尾鱼类样本的肠道原核生物和真核生物的多样性的曼-惠特尼U检验结果显示,鲢肠道内的原核生物的香浓多样性和物种数均显著低于鳙,而鲢真核生物的物种数要显著多于鳙。研究表明千岛湖鲢、鳙的摄食具有明显的差异。

In order to wnderstand the differences of the diets between Hypophthalmichthys molitrix and Aristichthys nobilis in Qiandao Lake, PCR-DGGE fingerprinting was adopted to analyze the variable regions of 16S rDNA of prokaryotes and 18S rDNA of eukaryotes.53 OTUs ( operational taxonomic units ) in prokaryotes and 45 OTUs in eukaryotes were ob-tained.Cluster analysis based on Dice similarity coefficient and principal components analysis ( PCA)of community indica -ted that intestinal content composition of H.molitrix and A.nobilis were slightly similar in prokaryotes communities and obvi-ously different in eukaryotes communities.Mann-Whitney U test of biodiversity and species number of intestinal contents of prokaryotes and eukaryotes between H.molitrix and A.nobilis showed that prokaryotes ’ Shannon diversity and species num-ber of H.molitrix were significantly lower than those of A.nobilis, while the number of eukaryotic species of H.molitrix was significantly more than that of A.nobilis.These results confirmed that H.molitrix and A.nobilis have different trophic niches in Qiandao Lake.
