Complement%Macrophages%Phagocytosis%Trichosporon asahii
目的研究补体对RAW264.7小鼠巨噬细胞吞噬阿萨希毛孢子菌(Trichosporon asahii, T.asahii)的增强作用。方法实验分为3组,实验组为T. asahii菌液加入10%正常小鼠血清37℃预孵育1 h。空白对照组为菌液不经正常小鼠血清预孵育。热灭活对照组为正常小鼠血清预先在56℃水浴箱孵育30 min,然后加入T. asahii菌液37℃预孵育30 min。3组预处理的T. asahii菌液加入RAW264.7巨噬细胞共培养30 min后在显微镜下观察RAW264.7细胞吞噬T. asahii的情况,计数吞噬个数及吞噬率。结果实验组与空白对照组及热灭活对照组的吞噬率比较,差异均具有显著性(P<0.01)。空白对照组与热灭活对照组的吞噬率比较,无显著性差异(P>0.05)。结论补体可增强RAW264.7小鼠巨噬细胞吞噬T. asahii的作用。

Objective To investigate the efficacy of complement on the phagocytosis activity of RAW264.7 macrophage against Trichosporon asahii (T. asahii). Methods T. asahii was preincubated with 10%normal mouse serum for 30 min at 37℃. In blank control group, T. asahii was not preincubated with 10%normal mice serum. In heat inactivation control group, normal mouse serum was incubated in water bath at 56℃for 30 min before co-incubated with T. asahii. After incubated with RAW264.7 macrophage for 30 min, the numbers of T. asahii cells phagocytosed by RAW264.7 macrophage were counted under an inverted microscope, and the rate of phagocytosis was calculated. Comparisons of parameters between these groups were done by ANOVA analysis. Results There were signiifcant difference between experimental group and the two control groups (P<0.01). No signiifcant difference was found between the two control groups (P>0.05). Conclusion Complement were able to enhance the phagocytosis of RAW264.7 macrophage to T. asahii.
