目的:探讨表皮生长因子(epidermal growth factor,EGF)和碱性成纤维生长因子(basic fibroblast growth factor, bFGF)对人牙髓干细胞(human dental pulp stem cells, hDPSCs)增殖能力的影响。方法:采用噻唑蓝(MTT)比色法分别测定不同浓度EGF和bFGF在不同时间,单独或联合应用对hDPSCs增殖能力的影响。结果:50 ng/ml EGF和20 ng/ml bFGF促hDPSCs增殖能力最强。以50ng/ml EGF、20ng/ml bFGF单独或联合作用,其促hDPSCs增殖能力均具有时间依赖性,第3 d、5 d、7 d, EGF、bFGF促hDPSCs增殖作用显著增强,二种生长因子联合作用,促增殖作用显著高于其单独作用效果,差异具有统计学意义(P<0.05)。结论:生长因子EGF和bFGF最大效应浓度分别为:50 ng/ml、20 ng/ml。EGF和bFGF单独作用均能显著促进hDPSCs的增殖,二者联合作用对hDPSCs的增殖有一定的协同作用。

Objective: To investigate the effects of epidermal growth factor (EGF) and basic fibroblast growth factor(bFGF)on the proliferation of human dental pulp stem cells(hDPSCs) in vitro. Methods: Colorimetric assay was used to measure the dose - and time - depedent effects of EGF and bFGF on the proliferation of hDPSCs. Results:50 ng/ml EGF and 20 ng/ml bFGF had the strongeste effect on promoting the proliferation of hDPSCs re-spectively. The effect of 50 ng/ml EGF and 20 ng/ml bFGF, whether alone or combined was time-depedent. It was significantly stronger at 3, 5, 7 d (P < 0.05), and the combined usege of the two cytokines was obviously stronger than used alone(P<0.05).Conclusion: The optimal concentration of EGF and bFGF is 50 ng/ml and 20 ng/ml respectively. Both EGF and bFGF can significantly promote the proliferation of hDPSCs. The combination use of two growth factors has a certain synergism.
