目的:用免疫组化联合放射自显影术观察移植骨髓间充质干细胞表达dystrophin蛋白情况。方法:用3H-TdR标记的骨髓间充质干细胞经静脉移植肌营养不良鼠(mdx),在1、2、3、4月取小鼠腓肠肌肉,先进行免疫组化染色,再将标本立即进行放射自显影,3月后再将组织HE染色,显微镜下观察干细胞是否表达了dystrophin蛋白。同时用激光共聚焦显微镜(LSCM)观察BrdU标记的间充质干细胞移植mdx鼠后的dystrophin蛋白表达为比较标准。结果:两法均观察到移植的干细胞表达了dystrophin蛋白,且观察到的阳性细胞数比较无统计学意义;干细胞表达dystrophin蛋白随着时间有逐渐增高趋势。结论:免疫组化联合放射自显影技术可以用于对干细胞移植后蛋白表达的示踪观察,干细胞移植对肌营养不良具有治疗前景。
目的:用免疫組化聯閤放射自顯影術觀察移植骨髓間充質榦細胞錶達dystrophin蛋白情況。方法:用3H-TdR標記的骨髓間充質榦細胞經靜脈移植肌營養不良鼠(mdx),在1、2、3、4月取小鼠腓腸肌肉,先進行免疫組化染色,再將標本立即進行放射自顯影,3月後再將組織HE染色,顯微鏡下觀察榦細胞是否錶達瞭dystrophin蛋白。同時用激光共聚焦顯微鏡(LSCM)觀察BrdU標記的間充質榦細胞移植mdx鼠後的dystrophin蛋白錶達為比較標準。結果:兩法均觀察到移植的榦細胞錶達瞭dystrophin蛋白,且觀察到的暘性細胞數比較無統計學意義;榦細胞錶達dystrophin蛋白隨著時間有逐漸增高趨勢。結論:免疫組化聯閤放射自顯影技術可以用于對榦細胞移植後蛋白錶達的示蹤觀察,榦細胞移植對肌營養不良具有治療前景。
목적:용면역조화연합방사자현영술관찰이식골수간충질간세포표체dystrophin단백정황。방법:용3H-TdR표기적골수간충질간세포경정맥이식기영양불량서(mdx),재1、2、3、4월취소서비장기육,선진행면역조화염색,재장표본립즉진행방사자현영,3월후재장조직HE염색,현미경하관찰간세포시부표체료dystrophin단백。동시용격광공취초현미경(LSCM)관찰BrdU표기적간충질간세포이식mdx서후적dystrophin단백표체위비교표준。결과:량법균관찰도이식적간세포표체료dystrophin단백,차관찰도적양성세포수비교무통계학의의;간세포표체dystrophin단백수착시간유축점증고추세。결론:면역조화연합방사자현영기술가이용우대간세포이식후단백표체적시종관찰,간세포이식대기영양불량구유치료전경。
Objective: To study dystrophin expression in mesenchymal stem cells from mdx mice by immunohistochemistry combined with autoradiography. Methods: Bone marrow mesenchymal stem cells labeled with 3H-TdR were transferred intravenously into 30 irradiated mdx mice 1、2、3 and 4 m after transfer, leg muscle of mdx rats was cryosected and fixed, then stained with routine immunohistochemical method 3 months later,it was stained by hematoxylin and eosin(HE), and observed by microscope. The other cryosections labeled by BrdU were observed by LSCM . Results: The distrophin expression of MSCs was detected by both methods without statistic difference.The expression increased gradually with the time. Conclusion: Dystrophin expression of MSCs can be observed by autoradiography combined with immunohistochemical technology. It may be applied to the evaluation of MSCs transplantation in myodystrophic patient in future.
