目的:通过丹花颗粒对急性心肌缺血大鼠炎症因子表达的影响,探讨其保护心肌缺血的作用机制,为进一步研究奠定基础。方法:120只Wistar大鼠,按照随机数字表法分为假手术组、模型组、丹花颗粒高、中、低剂量组、通心络组。灌胃14 d后,以结扎冠脉左前降支建立急性心肌缺血模型。全自动生化仪检测谷草转氨酶(AST)、乳酸脱氢酶(LDH)、肌酸酶同工酶(CK-MB);记录造模前后ST段变化;双抗夹心酶联免疫吸附方法(ELISA)检测白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、血栓素(TXA2)、前列环素(PGI2)、血清高敏c反应蛋白(CRP)、肌钙蛋白I(Tn-I)浓度;免疫组化方法检测心肌细胞NF-资B表达。结果:造模前后治疗组与模型组比较移ST变化显著(P<0.01)。与模型组比较,丹花颗粒高剂量组与通心络组AST、LDH、CK-MB、Tn-I浓度均明显降低(P<0.05),低剂量组与中剂量组CK-MB、Tn-I浓度明显降低(P<0.05),丹花颗粒组与通心络组CRP表达明显减少(P<0.05),丹花颗粒组与通心络组IL-6、TNF-α浓度明显降低(P<0.05),丹花颗粒组与通心络组PGI2水平均明显增加(P<0.05);TXA2水平明显降低(P<0.05)。丹花颗粒组及通心络组NF-资Bp65阳性表达明显减弱(P<0.05)。结论:丹花颗粒可能通过抑制NF-资B细胞外的正反馈激活途径,从而减少IL-6、TNF-α、TXA2、PGI2、CRP等炎症因子的基因转录,阻断后者所介导的一系列炎性反应,可能是其防治心肌缺血的作用机制之一。

목적:통과단화과립대급성심기결혈대서염증인자표체적영향,탐토기보호심기결혈적작용궤제,위진일보연구전정기출。방법:120지Wistar대서,안조수궤수자표법분위가수술조、모형조、단화과립고、중、저제량조、통심락조。관위14 d후,이결찰관맥좌전강지건립급성심기결혈모형。전자동생화의검측곡초전안매(AST)、유산탈경매(LDH)、기산매동공매(CK-MB);기록조모전후ST단변화;쌍항협심매련면역흡부방법(ELISA)검측백개소-6(IL-6)、종류배사인자-α(TNF-α)、혈전소(TXA2)、전렬배소(PGI2)、혈청고민c반응단백(CRP)、기개단백I(Tn-I)농도;면역조화방법검측심기세포NF-자B표체。결과:조모전후치료조여모형조비교이ST변화현저(P<0.01)。여모형조비교,단화과립고제량조여통심락조AST、LDH、CK-MB、Tn-I농도균명현강저(P<0.05),저제량조여중제량조CK-MB、Tn-I농도명현강저(P<0.05),단화과립조여통심락조CRP표체명현감소(P<0.05),단화과립조여통심락조IL-6、TNF-α농도명현강저(P<0.05),단화과립조여통심락조PGI2수평균명현증가(P<0.05);TXA2수평명현강저(P<0.05)。단화과립조급통심락조NF-자Bp65양성표체명현감약(P<0.05)。결론:단화과립가능통과억제NF-자B세포외적정반궤격활도경,종이감소IL-6、TNF-α、TXA2、PGI2、CRP등염증인자적기인전록,조단후자소개도적일계렬염성반응,가능시기방치심기결혈적작용궤제지일。
This study was aimed to explore mechanism of Danhua particles in the protection of myocardial ischemia , and lay the foundation for further study through the expression of inflammatory cytokines of Danhua particles in rats with acute myocardial ischemia . A total of 120 Wistar rats were divided into the sham group , model group, Danhua particles high-dose group, middle-dose group, low-dose group, Tongxinluo group, according to the random number table . The automatic biochemical analyzer was used in the detection of the concentration of the AST , LDH , CK-MB . And the ECG was used to record the ST-segment changes before and after modeling . The ELISA kit was used in the detection of concentration of IL-6 , TNF-α, TXA2 , PGI2 , CRP and TN-I . Immunohistochemical methods were used to detect myocardial cell NF-кB expression . The results showed that before and after the modeling , the treatment group compared with model group ∑ST changed significantly ( P < 0 . 01 ) . Compared with the model group , the Danhua particles high -dose group and Tongxinluo group were significantly reduced in the concentration of AST , LDH , CK-MB and Tn-I ( P <0 . 05 ); the low-dose group and middle-dose group were significantly reduced in the concentration of CK-MB and Tn-I ( P < 0 . 05 ); the expression of CRP in the Danhua particles group and Tongxinluo group was significantly reduced ( P < 0 . 05 ) . Danhua particles group and Tongxinluo group compared with the model group were significantly reduced in the concentration of IL-6 and TNF-α ( P < 0 . 05 ); the PGI2 level in the Danhua particles group and Tongxinluo group was significantly increased ( P < 0 . 05 ); and the TXA2 level was obviously
<br> reduced ( P < 0 . 05 ) . The positive expression of NF-кBp65 in the Danhua particles group and Tongxinluo group was significantly reduced ( P < 0 . 05 ) . It was concluded that Danhua particles inhibited the NF-кB extra cellular positive feedback activation pathway , and thus reduced the inflammatory factor gene transcription such as IL-6 , TNF-α, TXA2 , PGI2 , CRP , block a series of inflammatory reactions which were mediated . It may be one of the mechanisms of Danhua particles in the prevention on myocardial ischemia .
