目的研究肿瘤坏死因子-α( TNF-α)与声门不同状态下闭合性胸部撞击所致肺损伤的关系。方法采用逆转录聚合酶链反应( RT-PCR)法检测48只新西兰大白兔声门紧闭与(或)开放状态下,不同时相点伤侧肺组织TNF-αmRNA的表达,以6只正常新西兰大白兔为对照。结果在伤后2h时,声门紧闭与开放两组TNF-αmRNA的表达无显著差别(P>0.05),伤后4、8、12h声门紧闭组TNF-αmRNA的表达显著高于声门开放组( P<0.05)。伤侧肺组织中TNF-αmRNA表达各时相点显著高于对照组( P<0.05)。结论TNF-α参与创伤性肺损伤的发生发展并起重要作用,声门紧闭状态下肺撞击伤的损伤程度重于声门开放状态。
目的研究腫瘤壞死因子-α( TNF-α)與聲門不同狀態下閉閤性胸部撞擊所緻肺損傷的關繫。方法採用逆轉錄聚閤酶鏈反應( RT-PCR)法檢測48隻新西蘭大白兔聲門緊閉與(或)開放狀態下,不同時相點傷側肺組織TNF-αmRNA的錶達,以6隻正常新西蘭大白兔為對照。結果在傷後2h時,聲門緊閉與開放兩組TNF-αmRNA的錶達無顯著差彆(P>0.05),傷後4、8、12h聲門緊閉組TNF-αmRNA的錶達顯著高于聲門開放組( P<0.05)。傷側肺組織中TNF-αmRNA錶達各時相點顯著高于對照組( P<0.05)。結論TNF-α參與創傷性肺損傷的髮生髮展併起重要作用,聲門緊閉狀態下肺撞擊傷的損傷程度重于聲門開放狀態。
목적연구종류배사인자-α( TNF-α)여성문불동상태하폐합성흉부당격소치폐손상적관계。방법채용역전록취합매련반응( RT-PCR)법검측48지신서란대백토성문긴폐여(혹)개방상태하,불동시상점상측폐조직TNF-αmRNA적표체,이6지정상신서란대백토위대조。결과재상후2h시,성문긴폐여개방량조TNF-αmRNA적표체무현저차별(P>0.05),상후4、8、12h성문긴폐조TNF-αmRNA적표체현저고우성문개방조( P<0.05)。상측폐조직중TNF-αmRNA표체각시상점현저고우대조조( P<0.05)。결론TNF-α삼여창상성폐손상적발생발전병기중요작용,성문긴폐상태하폐당격상적손상정도중우성문개방상태。
Objective To study the relationship between TNF-αand lung injury induced by blunt chest trauma under different glottis states .Methods The TNF-αmRNA expression of the injured lung tissue in the 48 New Zealand rabbits was detected by reverse transcription polymerase chain reaction ( RT-PCR) under the closed and open glottis states at designed time .Meanwhile,6 healthy New Zealand rats were taken as control .Results There was no remarkable difference in the expressions of TNF-αof lung tissue between the closed glottis group and the open glottis group after 2 hours after injury(P>0.05).But the TNF-αmRNA expression was obviously higher in the closed glottis group than that in open glttis group at 4,8,and 12 hours after injury(P<0.05).TNF-αmRNA expressions were much higher in injured lung tissue than that in control group at designed time (P<0.05).Con-clusion TNF-αis involved in the occurrence and development of traumatic lung injuries and plays a critical role in this process .The severity of lung injuries under the closed glottis state is significantly higher than that under open glottis state .
