Cr(VI)-resistant bacterium%screening%identification%condition optimization
为了筛选对六价铬Cr(VI)有强耐性的细菌,采集铬污染土壤,通过稀释平板法进行初筛,将单菌落分离纯化,进一步将单菌落分别接种于含不同浓度的铬培养基上进行复筛,并通过形态特征和16S rDNA序列测定对Cr(VI)耐性较强的菌株进行鉴定.通过单因素试验考察pH、接种量、温度和培养时间对最优菌株耐Cr(VI)性能的影响.结果表明,初筛分离纯化出5株对Cr(VI)有一定耐性的菌株,命名为Cr1~Cr5.通过浓度梯度试验复筛出耐Cr(VI)能力较强的菌株Cr2,此菌株在含250 mg/L铬的培养基上生长良好,经鉴定为蜡样芽孢杆菌.该菌株的最佳培养条件为pH 7.0,4%Cr2种子液接种量,35℃培养3 d .在此条件下,对Cr(VI)的去除率达87.7%.该研究为微生物修复重金属污染土壤奠定了基础.

In order to isolate and screen Cr(VI)-resistant bacterium,chromium-contaminated soil were collected. Through the plate method for the first screening,five bacterial colonies were got,then these single colonies were isolated and purified,and then the single colonies were inoculated in LB medium containing different concentrations of chromium respectively for further screening. The high-efficiency Cr(VI)-resistant strain was identified by a series of bacterial colony morphological and 16S rDNA sequences. The effect of pH,inoculation amount,temperature and culture time on the efficiency of the optimum Cr(VI)-resistant strain were studied by single-factor test. Five Cr(VI)-resistant strains were isolated and purified,named Cr1~Cr5. Cr2,one high-efficiency Cr(VI)-resistant strain,was screened through chromium concentration gradient tests and it grew well in LB medium containing 250 mg/L chromium. Cr2 was identified as Bacillus cereus. The optimum culture condition was pH 7.0,4%inoculation amount,35 ℃ for 3 d. Under the condition,the chromium removal capacity of the bacteria was 87.7%. The study laid the fundation for the remediation of heavy metals-contaminated soil by microorganism.
