采用正交实验方法对聚焦微波辅助提取( FMAE)的实验条件进行优化,以天芥菜碱为提取目标物, HPLC法测定其含量。通过优化,得到最佳实验条件:药材粒度60目,液料比1:35,微波功率420 W,微波时间50 min。HPLC分析条件:以甲醇-0.05%三乙胺( pH 7.5)(30:70)为流动相,在220 nm处检测,天芥菜碱的线性范围为52~1040 mg/L,相关系数( r)为0.9999,检出限为0.0402 mg/L,其加标回收率为97.5%~103%,相对标准偏差为2.4%,方法简单、灵敏度高。

An extraction method of heliotrine was developed by focused microwave-assisted extraction ( FMAE). Orthogonal design was used to optimize the experimental conditions. The optimum extrac-tion conditions were as follows:60 mesh size of material,solvent-materail ratio:1 : 35,micro-wave power:420 W and extraction time:50 min. The analyte was determined by HPLC using meth-anol -0. 05% triethylamine(pH 7. 5 )(30 : 70)as mobile phase with a detection wavelength of 220 nm. The linear range of heliotrine was in the range of 52-1 040 mg/L(r=0. 999 9),with a detec-tion limit of 0. 040 2 mg/L. The recoveries for heliotrine were in the range of 97. 5% -103% with RSDs of 2. 4% . The method was simple and highly sensitive.
