目的:构建SATB1基因过表达的慢病毒载体,检测其在MCF-7乳腺癌细胞中的表达及对癌细胞侵袭性的影响。方法应用DNA重组技术,将SATB1基因插入到含有绿色荧光蛋白基因的慢病毒表达载体质粒GV287中,获得重组载体,经测序鉴定后转染293T细胞产生慢病毒载体GV287-SATB1,用GV287-SATB1转染人乳腺癌MCF-7,Western blot 分析转染前后SATB1表达情况,穿膜实验检测MCF-7细胞的侵袭性。结果成功构建人MCF-7细胞SATB1基因过表达慢病毒载体,MCF-7细胞转染慢病毒载体后SATB1蛋白表达水平显著上调,细胞的侵袭性显著增强。结论 SATB1基因过表达的慢病毒载体,可在MCF-7乳腺癌细胞中过表达SATB1蛋白,并显著增强MCF-7细胞的侵袭性。

목적:구건SATB1기인과표체적만병독재체,검측기재MCF-7유선암세포중적표체급대암세포침습성적영향。방법응용DNA중조기술,장SATB1기인삽입도함유록색형광단백기인적만병독표체재체질립GV287중,획득중조재체,경측서감정후전염293T세포산생만병독재체GV287-SATB1,용GV287-SATB1전염인유선암MCF-7,Western blot 분석전염전후SATB1표체정황,천막실험검측MCF-7세포적침습성。결과성공구건인MCF-7세포SATB1기인과표체만병독재체,MCF-7세포전염만병독재체후SATB1단백표체수평현저상조,세포적침습성현저증강。결론 SATB1기인과표체적만병독재체,가재MCF-7유선암세포중과표체SATB1단백,병현저증강MCF-7세포적침습성。
Objective To construct lentivirus overexpressed SATB 1,and to study its expression in MCF-7 cells and its effect on invasive breast cancer cells .Methods The SATB1 gene was inserted into lentivirus plasmid GV 287 and was detected by DNA sequencing.The recombined plasmid was transfected into 293T cell line to yield lentivirus GV287-SATB1,then the GV287-SATB1 was transfected into MCF-7 human breast cancer cells .The expression of SATB 1 was detected by Western bolt ,in-vasion of the cells was inspected by Transwell assay .Results The lentivirus overexpressed SATB 1 was correctly constructed . Transfection of GV287-SATB1 overexpressed SATB1 protein in MCF-7 cells and increased invasion of these cells .Conclusion The lentivirus overexpressed SATB 1 is successfully constructed .GV287-SATB1 can be overexpressed in MCF-7 cells and increase invasion of these cells .
