目的探讨荷包牡丹碱对人胃癌SGC-7901细胞生长的抑制作用及其可能的机制。方法体外培养人胃癌SGC-7901细胞，MTT法测定细胞增殖情况；Hoechst33258染色观察细胞凋亡形态学变化；West-ern blot法检测Bax、Bcl-2和Caspase-3的表达。结果 MTT法显示：不同浓度的荷包牡丹碱作用于胃癌SGC-7901细胞不同的时间之后，SGC-7901细胞呈现时间及浓度依赖性的生长抑制；流式细胞仪方法显示：荷包牡丹碱可以促进胃癌SGC-7901细胞的凋亡；行Hoechest33258染色可见细胞核出现固缩、边集，形成凋亡小体等凋亡现象；Western blot法检测显示：与对照组相比，药物作用后Bcl-2表达降低，Bax、Caspase-3表达升高， Bcl-2/Bax比例失调。结论荷包牡丹碱在体外对胃癌细胞的增殖与生长具有抑制作用，可促进其凋亡，其作用可能与Bcl-2/Bax降低导致Caspase-3活化有关。

Objective To investigate the effect of Dicentrine on the human gastric carcinoma cell SGC-7901 and the possible mechanisms .Methods SGC-7901 gastric cancer cells were cultured in a controlled environment . Examining the inhibitory effect on proliferation of cells ,which were measured by MTT assay .Morphological changes of cells were observed by Hoechest 33258 staining.The expression of Bax , Bcl-2 and Caspase-3 were detected by Western-blot.Results The results acquired from the MTT assay showed that the proliferation of SGC-7901 cells were inhibited significantly by the dicentrine .The results showed that an increase in concentration corresponded directly with the length of time it took for the SGC-7901 cells to culture,with a higher concentration resulting in a stunt in cell growth .Apoptotic features such as nuclear pyknosis , chromatin margination , and apoptotic bodies were observed by Hoechest33258 staining.Compared with the control group ,the expression of Bcl-2 was decreased while that of Bax and Caspase-3 were increased in the group of dicentrine .Conclusions Dicentrine has a significant effect on the inhibition of gastric carcinoma cells and the apoptosis was promoted .The effect may be mediated by down-regulating Bcl-2/Bax,resulting in Caspase-3 activition.
