目的观察蝎毒增殖多肽( Buthus martensii Karsch proliferation peptide ,BmKpp)对X射线辐射后大鼠骨髓间充质干细胞(rat bone-marrow-derived mesenchymal stem cells ,rBMSCs)生长的影响。方法利用全骨髓细胞贴壁法分离纯化rBMSCs,体外扩增传代,取第3代至第8代细胞为实验对象。流式细胞术对rBMSCs进行细胞表型鉴定后,将细胞设为非辐射组( IR-组)和辐射组( IR+组,总剂量为8.0 Gy、标称剂量率300 MU/min的X射线照射)。 CCK-8法检测BmKpp对各组细胞在不同时间的增殖情况。结果体外培养的第3代或第8代rBMSCs 的细胞表型均为:CD29+/CD90+/CD34-/CD45-。在BmKpp对X射线辐射后rBMSCs作用的最低有效浓度筛选的实验中,BmKpp 8μg/ml处理组的吸光度( A值)显著高于辐射对照组( P<0.01);BmKpp(8μg/ml)有促进辐射后的rBMSCs增殖的作用,尤以辐射后第8天、第16天、第18天和第20天效果最为明显,其A值与对照组的相比,P均<0.05;BmKpp(8μg/ml)未能显著促进非辐射rBMSCs的增殖,与对照组相比无统计学差异( P>0.05)。结论 BmKpp能促进辐射后的rBMSCs的增殖,从而发挥其促进辐射后造血功能的恢复作用。

목적관찰갈독증식다태( Buthus martensii Karsch proliferation peptide ,BmKpp)대X사선복사후대서골수간충질간세포(rat bone-marrow-derived mesenchymal stem cells ,rBMSCs)생장적영향。방법이용전골수세포첩벽법분리순화rBMSCs,체외확증전대,취제3대지제8대세포위실험대상。류식세포술대rBMSCs진행세포표형감정후,장세포설위비복사조( IR-조)화복사조( IR+조,총제량위8.0 Gy、표칭제량솔300 MU/min적X사선조사)。 CCK-8법검측BmKpp대각조세포재불동시간적증식정황。결과체외배양적제3대혹제8대rBMSCs 적세포표형균위:CD29+/CD90+/CD34-/CD45-。재BmKpp대X사선복사후rBMSCs작용적최저유효농도사선적실험중,BmKpp 8μg/ml처리조적흡광도( A치)현저고우복사대조조( P<0.01);BmKpp(8μg/ml)유촉진복사후적rBMSCs증식적작용,우이복사후제8천、제16천、제18천화제20천효과최위명현,기A치여대조조적상비,P균<0.05;BmKpp(8μg/ml)미능현저촉진비복사rBMSCs적증식,여대조조상비무통계학차이( P>0.05)。결론 BmKpp능촉진복사후적rBMSCs적증식,종이발휘기촉진복사후조혈공능적회복작용。
Objective To study the effect of Buthus martensii Karsch proliferation peptide ( BmKpp) on rat bone-marrow-derived mesenchymal stem cells ( rBMSCs ) growth after X-ray radiation.Methods rBMSCs were isolated from the whole bone marrow and cultured in vivo.Take P3-P8 cells for experimental subject .Cellular immune phenotype was identified by flow cytomertry .rBMSCs were divided into two experimental groups:normal group ( non-radiation group ,IR-) ,and irradiation group ( IR+) which rBMSCs were exposed to X-ray for 8.0 Gy total dose and nominal dose rate 300 MU/min with SAD method .Each experimental group was composed of control group and BmKpp treated groups .CCK-8 method was used to examine the proliferation of rBMSCs of different groups at different times.Results The cellular immune phenotype identification of P 3-P8 cells was showed as CD29 +/CD90 +/CD34 -/CD45 -.The absorbance value of 8 μg/ml BmKpp treated group was higher significantly than that of control group(P<0.01).BmKpp(8 μg/ml)could promote irradiated rBMSCs proliferation effectively ,especially at the 8th day,16th day,18th and 20th day after irradiation(P <0.05).But BmKpp(8 μg/ml)could not promoting the proliferation of non-irradiated rBMSCs .Conclusions BmKpp could promote X-ray irradiated rBMSCs proliferation , but has no obvious effect on normal rBMSCs .It indicated that BmKpp promote the proliferation of rBMSCs , which need to be in a certain stress conditions ( eg.irradiation ) , may be related to the recovery of hematopoietic function after irradiation.
