为寻找松江鲈( Trachidermus fasciatus)鸭绿江种群( YL)、黄河种群( YR)和富春江种群( FR)间的分子标记,从120条10 bp随机引物中,筛选出多态性引物54条进行RAPD扩增。克隆获得了S2069680 bp、 S2082331 bp、 S2105782 bp与S2111400 bp等4个清晰的 RAPD 特异性标记,并成功将 RAPD 标记 S2082331 bp与 S2105782 bp转化成 SCAR03260 bp和SCAR04710 bp的SCAR标记。经每群体30尾样本验证,发现SCAR03260 bp在YL、 YR和FR种群中出现的频率依次为13.33%、93.33%、86.67%,可作为鉴别鸭绿江种群与其它两种群的分子标记;相反, SCAR04710 bp在YL、YR种群的出现频率较高(86.67%、83.33%),而在FR种群的出现频率却很低(16.67%),可作为鉴别富春江种群与其它两种群的分子标记。

To search for molecular markers among the Yalu River population (YL), Yellow River population (YR) and Fuchun River population ( FR) of Trachidermus fasciatus, fifity-four polymorphic 10-bp random primers were selected from 120 ones and used in the RAPD analysis.In total, four clear specific RAPD markers , S2069 680 bp , S2082 331 bp , S2105 782 bp and S2111 400 bp were obtained.But only S2082 331 bp and S2105 782 bp were successfully transformed into SCAR markers (SCAR03260 bp and SCAR04710 bp, respectively).Afterwards, the frequencies of the two SCAR makers were tested in 30 samples for each population.As result, the range of frequency for SCAR 03260 bp was 13.33%, 93.33% and 86.67% in YL, YR and FR population respectively.Conversely, the frequencies of SCAR04710 bp in YL and YR populations were 86.67%and 83.33% respectively, which were obviously higher than that in FR (16.67%).Hence, SCAR03260 bp or SCAR04710 bp can be used as special molecular marker for the population identification between YL and YR , FR, or be-tween FR and YR, YR, respectively.
