目的:探讨体外感染减毒麻疹病毒(MV)对子宫内膜腺癌细胞(JEC)的凋亡诱导作用及相关机理。方法:MTT比色法动态监测MV感染对JEC细胞增殖活力的影响;琼脂糖凝胶电泳以及流式细胞术检测细胞凋亡;免疫细胞化学法检测MV感染后JEC细胞的Fas、FasL及TGF-β表达,分析凋亡机制。结果:MV感染6 d内,促进细胞的生长;感染6 d后,抑制细胞的生长,最终造成细胞死亡。流式细胞术结果显示:各感染浓度组JEC细胞的凋亡率比正常对照组增高(P<0.05或P<0.01),随感染时间的延长,细胞的凋亡率呈上升的趋势,感染12 d时,凋亡率与感染浓度呈正相关(r=0.77),且琼脂糖凝胶电泳出现特征性DNA“Ladder”凋亡条带;此时,感染细胞内Fas及TGF-β的表达增强(P<0.01),而FasL始终未表达。结论:MV感染能够抑制JEC细胞增值,并诱导其凋亡,JEC细胞凋亡机制可能与Fas、TGF-β的表达有关。

목적:탐토체외감염감독마진병독(MV)대자궁내막선암세포(JEC)적조망유도작용급상관궤리。방법:MTT비색법동태감측MV감염대JEC세포증식활력적영향;경지당응효전영이급류식세포술검측세포조망;면역세포화학법검측MV감염후JEC세포적Fas、FasL급TGF-β표체,분석조망궤제。결과:MV감염6 d내,촉진세포적생장;감염6 d후,억제세포적생장,최종조성세포사망。류식세포술결과현시:각감염농도조JEC세포적조망솔비정상대조조증고(P<0.05혹P<0.01),수감염시간적연장,세포적조망솔정상승적추세,감염12 d시,조망솔여감염농도정정상관(r=0.77),차경지당응효전영출현특정성DNA“Ladder”조망조대;차시,감염세포내Fas급TGF-β적표체증강(P<0.01),이FasL시종미표체。결론:MV감염능구억제JEC세포증치,병유도기조망,JEC세포조망궤제가능여Fas、TGF-β적표체유관。
Objective:To study the apoptosis-inducing effects and mechanism of infection with measle vaccinal virus on adenocarcinoma endometrium cell lines(JEC)in vitro.Method:The proliferation abilities of JEC cells affected by measle vaccinal virus was examined by MTT;the effects of apoptosis were detected using the following methods:DNA agarose gel electrophoresis,Annexin-V FITC/PI stained FCM;the expressions of Fas, FasL and TGF-βof JEC were examined by immunocytochemical staining.Result:(1)After infection,the results of MTT showed MV promoted the proliferation abilities of JEC cells in 6 days,then inhibited,at last induced the JEC cells to death after 6th day.And FITC/PI stained FCM showed that the percentage of the JEC apoptotic cells were higher than the negative control groups(P<0.05),and the apoptotic rate had the increasing tendency following with the prolonging of infected time;furthermore,at 12 day,the concentrations of virus were positive correlation with the apoptotic rate(r=0.77),at the same time,ladder-like pattern of DNA fragments were demonstrated by agarose gel electrophoresis. Simultaneously,The results of immunocytochemical staining method showed that the expression of Fas and TGF-βrose after infected with MV in JEC(P<0.01),but FasL had no expression always. Conclusion:These results indicate that measle vaccinal virus can inhibited the proliferation abilities and induce apoptosis in JEC cell line in vitro;the apoptotic mechanism may be associated with the expressions of Fas and TGF-β.
