Type 2 diabetes%rosiglitazon%PDGF%implant osseointegration
目的:探讨罗格列酮对2型糖尿病大鼠种植体周围血小板衍生生长因子-B (PDGF-B)的影响。方法:选用2月龄SD大鼠30只,随机分为正常种植组(G1)、糖尿病种植组(G2)和糖尿病种植罗格列酮干预组(G3)。糖尿病模型采用高糖高脂饮食加小剂量链脲佐菌素诱导建立;种植模型于大鼠胫骨近骺端植入1.3mm×5mm纯钛种植体;罗格列酮干预组每日灌胃给药。三组大鼠每周测定一次血糖,于12周时经双能X线骨密度仪测定大鼠全身骨密度,处死大鼠后收集右侧胫骨,胫骨经固定、脱钙后制成切片,常规苏木精-伊红染色( HE染色),免疫组织化学法检测种植体周围骨组织中血小板衍生生长因子-B的表达情况。结果:正常组大鼠种植体周围血小板衍生生长因子-B的表达较多,糖尿病组次之,罗格列酮干预组表达最少;全身骨密度值的测量结果与免疫组化结果一致。结论:罗格列酮虽能降低2型糖尿病大鼠的血糖,但其对糖尿病大鼠骨组织有加重损害作用,其作用机制可能是通过抑制骨组织中血小板衍生生长因子-B的表达而完成的。
目的:探討囉格列酮對2型糖尿病大鼠種植體週圍血小闆衍生生長因子-B (PDGF-B)的影響。方法:選用2月齡SD大鼠30隻,隨機分為正常種植組(G1)、糖尿病種植組(G2)和糖尿病種植囉格列酮榦預組(G3)。糖尿病模型採用高糖高脂飲食加小劑量鏈脲佐菌素誘導建立;種植模型于大鼠脛骨近骺耑植入1.3mm×5mm純鈦種植體;囉格列酮榦預組每日灌胃給藥。三組大鼠每週測定一次血糖,于12週時經雙能X線骨密度儀測定大鼠全身骨密度,處死大鼠後收集右側脛骨,脛骨經固定、脫鈣後製成切片,常規囌木精-伊紅染色( HE染色),免疫組織化學法檢測種植體週圍骨組織中血小闆衍生生長因子-B的錶達情況。結果:正常組大鼠種植體週圍血小闆衍生生長因子-B的錶達較多,糖尿病組次之,囉格列酮榦預組錶達最少;全身骨密度值的測量結果與免疫組化結果一緻。結論:囉格列酮雖能降低2型糖尿病大鼠的血糖,但其對糖尿病大鼠骨組織有加重損害作用,其作用機製可能是通過抑製骨組織中血小闆衍生生長因子-B的錶達而完成的。
목적:탐토라격렬동대2형당뇨병대서충식체주위혈소판연생생장인자-B (PDGF-B)적영향。방법:선용2월령SD대서30지,수궤분위정상충식조(G1)、당뇨병충식조(G2)화당뇨병충식라격렬동간예조(G3)。당뇨병모형채용고당고지음식가소제량련뇨좌균소유도건립;충식모형우대서경골근후단식입1.3mm×5mm순태충식체;라격렬동간예조매일관위급약。삼조대서매주측정일차혈당,우12주시경쌍능X선골밀도의측정대서전신골밀도,처사대서후수집우측경골,경골경고정、탈개후제성절편,상규소목정-이홍염색( HE염색),면역조직화학법검측충식체주위골조직중혈소판연생생장인자-B적표체정황。결과:정상조대서충식체주위혈소판연생생장인자-B적표체교다,당뇨병조차지,라격렬동간예조표체최소;전신골밀도치적측량결과여면역조화결과일치。결론:라격렬동수능강저2형당뇨병대서적혈당,단기대당뇨병대서골조직유가중손해작용,기작용궤제가능시통과억제골조직중혈소판연생생장인자-B적표체이완성적。
Objective: To investigate the effects of Rosiglitazon on PDGF-B in osseous tissue around implant of type 2 diabetic rats. Methods: Thirty two-month-old rats were randomly divided into three groups:normal implant group (G1), diabetic implant group (G2) and diabetic implant group with Rosiglitazon intervention (G3). The diabetic modal was founded by feeding high-glucose-high-fat diet and injecting low dose streptozotocin (STZ). The implant modal was founded by inserting the titanium implant whose diameter is 1.3mm and length is 5mm in the right tibia near the epiphseal end. The intervention group was fed Rosiglitazon by rat`s stomach. 12 weeks later, the rats bone mineral density were measured, then the rats were killed and taken the right tibia. The hard tissue was fixed, decalcified and sectioned with conventional HE staining and immunohistochemistry to detect the expression of PDGF-B around implant. Results: The PDGF-B expression of normal is intense, PDGF-B expression of diabetic group is less, but, the intervention group is the least. The results of bone mineral density are consistent with the results of immunohistochemistry. Conclusion: Rosiglitazon can decrease blood glucose of diabetic rat, however, is bad for the bone tisse, its mechanism is likely to suppress the PDGF-B expression of bone tisse to achieve the perniciasm.
