目的:观察外置磁场能否提高超顺磁性氧化铁(SPIO )标记的脂肪干细胞经静脉移植治疗SD大鼠急性脑缺血的成功率。方法:线栓法制备MCAo大鼠模型40只,随机分成缺血未干预对照组(IO组)、未磁性标记ADSCs静脉移植组(IC组),磁性标记ADSCs静脉移植组(ICS组)和磁性标记ADSCs静脉移植并加外加磁场组(ICSM组),各组在细胞移植后1天,7天分别进行行为学评分,普鲁士蓝染色观察磁标记ADSCs在大脑内的分布情况,TTC染色比较缺血体积,原子吸收光谱仪测各组大脑铁含量。结果:细胞移植后7天ICSM 组动物较其他三组神经系统功能明显改善( P<0.05),ICSM组缺血区普鲁士蓝染色阳性细胞增多,缺血体积显著减小( P<0.01),铁含量显著增多( P<0.01)。结论:外磁场可以诱导SPIO标记的ADSCs在体内的定向迁移,提高尾静脉移植ADSCs治疗急性脑缺血大鼠的成功率。

목적:관찰외치자장능부제고초순자성양화철(SPIO )표기적지방간세포경정맥이식치료SD대서급성뇌결혈적성공솔。방법:선전법제비MCAo대서모형40지,수궤분성결혈미간예대조조(IO조)、미자성표기ADSCs정맥이식조(IC조),자성표기ADSCs정맥이식조(ICS조)화자성표기ADSCs정맥이식병가외가자장조(ICSM조),각조재세포이식후1천,7천분별진행행위학평분,보로사람염색관찰자표기ADSCs재대뇌내적분포정황,TTC염색비교결혈체적,원자흡수광보의측각조대뇌철함량。결과:세포이식후7천ICSM 조동물교기타삼조신경계통공능명현개선( P<0.05),ICSM조결혈구보로사람염색양성세포증다,결혈체적현저감소( P<0.01),철함량현저증다( P<0.01)。결론:외자장가이유도SPIO표기적ADSCs재체내적정향천이,제고미정맥이식ADSCs치료급성뇌결혈대서적성공솔。
Objective :The study aims to label adipose -derived stem cells (ADSCs) with superparamagnetic iron oxide ,then observe whether the magnetic field could improve the success rate of ADSCs intravenous transplantation in the treatment of acute cerebral ischemia in SD rats .Method :A total of 40 SD rat middle cerebral artery occlusion (MCAO ) models were established .Models were separated into four groups :ischemia only (IO ) ,ischemia with injection of cells (IC ) .ischemia with injection of SPIO -labeled cells (ICS ) ,and ischemia with injection of labeled cells and magnet exposure (ICSM ) .Twenty -four hours after middle cerebral artery occlusion (MCAo) ,labeled Rats ADSCs were injected into the tail vein .At one and Seven days after MCAo ,The neurological severity score (NSS ) was used to evaluate neurological function recovery of rats following the transplantation .The transplanted ADSCs were analyzed in recipient rat brains by Prussian blue staining .The infarct volume of rats were analyzed by TTC staining .The iron content of rat brains was determined by atomic absorption furnace spectrophotometry .Results :Significant improvement in NSS was observed in rats of ICSM group compared with the other groups after 7 days (P<0 .05 ) .Blue staining of brain slices showed a higher number of Prussian blue -positive ADSCs in brains of ICSM group than in brains of ICS group . Comparison of iron concentration in the ipsilateral hemisphere between the ICS and ICSM groups showed that there was a significantly higher iron concentration in the latter ( p<0 . 01) .TTC staining showed that infarct volume in ICSM rats was significantly reduced (P<0 .01) .Conclusion :External magnetic field can induce adipose -derived stem cells labeled with superparamagnetic iron oxide to migrate with target in vivo ,use of a magnetic field may be useful for improving the efficacy of targeted migration of stem cells in stem -based cell therapy in ischemic brain injury .
