目的:研究双氢青蒿素(Dihydroartemisinin ,DHA )对人骨肉瘤143B细胞裸鼠移植瘤肺转移的抑制作用及机制探讨。方法:将人骨肉瘤细胞143B接种于Bblb/c裸鼠股骨骨膜下,建立骨肉瘤模型。再将造模成功裸鼠随机分配为5组,即模型对照组、溶剂对照组和实验组(5、20、40 mg/kg DHA )。动态观察各组裸鼠生存状况和肿瘤体积变化,实验持续5周,过量麻醉法处死全部裸鼠,测量瘤重、计算瘤重抑制率,测量体积,计算肿瘤体积抑制率。肿瘤组织病理学分析。结果:5、20、40mg/kg DHA组肿瘤体积抑制率分别为7.1%、31.09%和72.89%,瘤重抑制率分别为12.12%、37.78%和55.0%,DHA处理组肿瘤体积和肿瘤重量与对照组比较,差异有统计学意义(P<0.05)。肿瘤组织病理学分析显示为典型的人骨肉瘤组织,组织学特征明显;且肺组织病理学分析结果显对照组裸鼠肺组织可见有典型的人骨肉瘤转移灶,而DHA处理组未发现有可见的人骨肉瘤转移灶。结论:双氢青蒿素不仅具有较强的抗人骨肉瘤活性,而且对抑制肺转移也具有显著的效果。

목적:연구쌍경청호소(Dihydroartemisinin ,DHA )대인골육류143B세포라서이식류폐전이적억제작용급궤제탐토。방법:장인골육류세포143B접충우Bblb/c라서고골골막하,건립골육류모형。재장조모성공라서수궤분배위5조,즉모형대조조、용제대조조화실험조(5、20、40 mg/kg DHA )。동태관찰각조라서생존상황화종류체적변화,실험지속5주,과량마취법처사전부라서,측량류중、계산류중억제솔,측량체적,계산종류체적억제솔。종류조직병이학분석。결과:5、20、40mg/kg DHA조종류체적억제솔분별위7.1%、31.09%화72.89%,류중억제솔분별위12.12%、37.78%화55.0%,DHA처리조종류체적화종류중량여대조조비교,차이유통계학의의(P<0.05)。종류조직병이학분석현시위전형적인골육류조직,조직학특정명현;차폐조직병이학분석결과현대조조라서폐조직가견유전형적인골육류전이조,이DHA처리조미발현유가견적인골육류전이조。결론:쌍경청호소불부구유교강적항인골육류활성,이차대억제폐전이야구유현저적효과。
Objective To study dihydroartemisinin (DHA ) on human osteosarcoma 143B cells in nude mice lung metastasis inhibition and its mechanism . Methods 143B human osteosarcoma cells were seeded in Bblb/c nudes’ mice periosteum ,the establishment of osteosarcoma model .Then successful modeling nude mice were randomly assigned to five groups ,namely ,model control group ,solvent control group and the experimental group (5 ,20 ,40 mg/kg DHA ) .Dynamic observation of the living conditions of nude mice in each group and tumor volume change ,experiment lasted five weeks ,all the mice were sacrificed by anesthetic overdose , measuring tumor weight ,tumor weight inhibition rate calculation , and measure the volume , tumor volume was calculated inhibition rate . Histopathological analysis .Results 5、20、40μg/kg DHA groups were suppressed tumor volume 7 .1% 、31 .09% and 72 .89% ,tumor weight inhibition rates were 12 .12% 、37 .78% and 55 .0% ,DHA treated tumor volume and tumor weight compared with the control group ,the difference was statistically significant (P <0 . 05) .Histopathological analysis of the tumor showed typical human osteosarcoma tissues ,histological characteristics significantly ;and lung histopathology analysis of lung tissue in nude mice in the control group was seen with typical human osteosarcoma metastases ,while DHA -treated group showed no visible human osteosarcoma metastases .Conclusion Dihydroartemisinin not only has a strong anti -tumor activity in human osteosarcoma ,and on the inhibition of lung metastasis also has a significant effect .
