目的:检测哮喘患儿外周血髓系抑制细胞(MDSCs)、白细胞介素-10(IL-10)和白细胞介素-12(IL-12)的水平。方法:应用两色荧光标记技术流式细胞仪检测哮喘发作组(n=52)、哮喘缓解组(n=50)、肺炎对照组(n=45)及正常对照组(n=48)儿童外周血MDSCs,用双抗体夹心ELISA法检测血清IL-10、IL-12水平。结果:哮喘发作组较其他3组外周血 MDSCs、血清IL-10水平升高,血清IL-12水平降低,差异有统计学意义( F=1043.820、1284.470和1139.380,P均<0.05);肺炎对照组与正常对照组各指标差异无统计学意义(P>0.05)。哮喘发作组患儿外周血MDSCs水平与IL-10呈正相关(r=0.741,P=0.007),与IL-12呈负相关(r=-0.879,P=0.014)。结论:MDSCs可能通过上调IL-10、下调IL-12在儿童哮喘发病中起作用。

목적:검측효천환인외주혈수계억제세포(MDSCs)、백세포개소-10(IL-10)화백세포개소-12(IL-12)적수평。방법:응용량색형광표기기술류식세포의검측효천발작조(n=52)、효천완해조(n=50)、폐염대조조(n=45)급정상대조조(n=48)인동외주혈MDSCs,용쌍항체협심ELISA법검측혈청IL-10、IL-12수평。결과:효천발작조교기타3조외주혈 MDSCs、혈청IL-10수평승고,혈청IL-12수평강저,차이유통계학의의( F=1043.820、1284.470화1139.380,P균<0.05);폐염대조조여정상대조조각지표차이무통계학의의(P>0.05)。효천발작조환인외주혈MDSCs수평여IL-10정정상관(r=0.741,P=0.007),여IL-12정부상관(r=-0.879,P=0.014)。결론:MDSCs가능통과상조IL-10、하조IL-12재인동효천발병중기작용。
Aim:To investigate the percentage of myeloid derived suppressor cells (MDSCs) and interleukin-10(IL-10), interleukin-12(IL-12) levels in peripheral blood from children with asthma .Methods: Taking 48 cases of healthy children and 45 cases of pneumonia children as control , the percentage of MDSCs in peripheral blood of 52 cases of atopic asthma and 50 cases of alleviated asthma were measured by double-color fluorescence labeling technique flow cytometry ,and the IL-10 and IL-12 levels in serum were measured by double antibody sandwich ELISA .Results:The percentage of MD-SCs and the serum from children with atopic asthma were significantly higher than those in other 3 groups, while the IL-12 levels in serum from children with atopic asthma was significantly lower (F=1 043.820,1 284.470,1 139.380,P<0.05). The difference between pneumonia and healthy control group was not obvious (P>0.05).In children with atopic asthma, the percentage of MDSCs had positive correlation with IL-10 level(r=0.741,P=0.007), while had negative correlation with IL-12(r=-0.879,P=0.014).Conclusion:MDSCs may play an important role in asthma through up-regulating IL-10 level, and down-regulating IL-12 level.
