石参为百合科独尾草属独尾草( Eremurus chinensis Fedtsch .)肉质根加工的稀有山野菜,主产于秦岭南坡局地。通过单因素和正交试验,研究了苯酚-硫酸法测定石参多糖含量时各因素对结果的影响,并对这些因素进行了优化和方法学验证。研究结果表明,苯酚用量1.6 mL、浓硫酸用量7.0 mL、显色时间30 min、显色温度80℃为苯酚-硫酸法测定石参多糖的最佳工艺条件,在此条件下平均回收率为98.64%,相对标准偏差( RSD )为1.40%;优化后的苯酚-硫酸法测定石参多糖准确性和稳定性高。比较分析表明,不同植物多糖的苯酚-硫酸法检测工艺有一定的差异,浓硫酸的加入方式对测定结果有较大影响。

Shishen , a rare edible wild herb produced in Qinling Mountains , is the process fleshy roots from Eremurus chinensis belonging to genus of Eremurus, Liliaceae.Through the single factor experiments and orthogonal experiments , the factors affecting the determination of polysaccharide content in Shishen ( E. chinensis) with phenol-sulfuric acid method were optimized .The results showed that the optimum determina-tion conditions were 1.6 mL phenol, 7.0 mL sulphuric acid, 80 ℃reaction temperature and 30 min reaction time.The average recovery was up to 98.64%, and the RSD was 1.40%.The comparative analysis indicated that the way used with phenol-sulfuric acid and how to add sulphuric acid maybe result in a great of influence to the results for different polysaccharides test .In addition , the modified phenol-sulfuric acid method to deter-minate polysaccharides is accurate and steady .
