目的探讨泡状棘球蚴感染对大鼠肝移植物存活的影响及其可能的机制。方法建立BN大鼠泡状棘球蚴感染模型,将实验大鼠分为实验组(LEW大鼠→感染泡球蚴3个月的BN大鼠)和对照组(健康LEW大鼠→健康BN大鼠),利用双袖套发建立肝移植模型,分别在移植术后第1、3、5、7天取肝脏组织,采用免疫组织化学的方法检测肝移植后排斥反应;采用流式细胞仪检测外周血中T淋巴细胞标志物CD4+、CD8+、CD28+的表达,利用半定量RT-PCR的方法检测趋化因子Fractalkine(FKN)的表达。结果实验组移植物存活时间[(15.5±3.93)d]明显长于对照组[(4.75±1.58)d],差异有统计学意义(P<0.05);免疫组化结果示实验组排斥反应较对照组出现较晚且程度较轻;实验组CD4+、CD8+及CD28+的表达[(41.35±4.58)%、(21.94±2.61)%、(17.76±4.16)%]较对照组[(51.28±6.36)%、(33.85±3.02)%、(23.08±4.74)%]低,差异有统计学意义(P<0.05);实验组FKNmRNA表达较对照组降低,差异有统计学意义(P<0.05)。结论泡状棘球蚴感染能够延长大鼠肝移植物的存活,泡状棘球蚴感染对宿主免疫系统尤其对效应T淋巴细胞进行抑制是可能的途径之一。

목적탐토포상극구유감염대대서간이식물존활적영향급기가능적궤제。방법건립BN대서포상극구유감염모형,장실험대서분위실험조(LEW대서→감염포구유3개월적BN대서)화대조조(건강LEW대서→건강BN대서),이용쌍수투발건립간이식모형,분별재이식술후제1、3、5、7천취간장조직,채용면역조직화학적방법검측간이식후배척반응;채용류식세포의검측외주혈중T림파세포표지물CD4+、CD8+、CD28+적표체,이용반정량RT-PCR적방법검측추화인자Fractalkine(FKN)적표체。결과실험조이식물존활시간[(15.5±3.93)d]명현장우대조조[(4.75±1.58)d],차이유통계학의의(P<0.05);면역조화결과시실험조배척반응교대조조출현교만차정도교경;실험조CD4+、CD8+급CD28+적표체[(41.35±4.58)%、(21.94±2.61)%、(17.76±4.16)%]교대조조[(51.28±6.36)%、(33.85±3.02)%、(23.08±4.74)%]저,차이유통계학의의(P<0.05);실험조FKNmRNA표체교대조조강저,차이유통계학의의(P<0.05)。결론포상극구유감염능구연장대서간이식물적존활,포상극구유감염대숙주면역계통우기대효응T림파세포진행억제시가능적도경지일。
Objective To study the technique and complication of orthotopic liver transplantation with E .m infected rats and observe the immune rejection of liver transplantation .Methods 24 Lew rats which were the recipient were randomly assigned to two groups .Group A was transplanted with the infected Lew re-cipient (the Lew were infected E .m 3 monthes later) .Group B was transplanted group with the uninfected Lew recipient .“Two-cuff method”is used to build the model .The liver tissue is taken on the 1st ,3rd ,5th , and 7th day after the transplantation .Immunohistochemistry method was used to detect liver transplanta-tion rejection ;using flow cytometry to test the expression of T lymphocyte markers CD4+ ,CD8+ ,CD28+in peripheral blood ;the use of semi-quantitative RT-PCR method to detect chemokine Fractalkine (FKN ) expression .Results The survival time of group A was (15 .50 ± 3 .93) days while the control group (4 .75 ± 1 .58) days ,and the difference was statistically significant (P < 0 .05);immunohistochemical results show rejection of the experimental group is later and milder than in the control group ;in the experimental group expression of CD4+ ,CD8+ and CD28+ [(41 .35 ± 4 .58)% ,(21 .94 ± 2 .61)% ,(17 .76 ± 4 .16)% ] is lower than the control group [(51 .28 ± 6 .36)% ,(33 .85 ± 3 .02)% ,(23 .08 ± 4 .74)% ] ,and the difference is statistically significant (P <0 .05);FKN mRNA expression gets lower in the experimental group than the control group ,the difference is statistically significant .Conclusion Survival time of Lew with AE was prominently longer than normal Lew .We will get new way to study liver transplantation with infected re-cipient and to study the tolerance of liver transplantation .The model can be a new choice for the immuno-logical study of live transplantation .
