目的比较白芍、赤芍水提物中所含化学成分的异同,及探讨白芍、赤芍水提物作用于体外大鼠胸动脉平滑肌株(A7r5)的生物效应差异。方法通过HPLC及质谱分析,比较白芍、赤芍水提物中各单体化学成分的共性与差异。采用体外培养A7r5,利用血小板源生长因子BB(PDGF-BB)诱导A7r5建立细胞增殖病理模型,加入不同剂量的白芍、赤芍水提物,通过MTT比色法和实时细胞分析仪检测细胞活性。结果白芍、赤芍在化学成分上有差别,即使两者拥有共同的成分,含量亦存在差异。在A7r5增殖研究显示,300μg/ml白芍水提液与A7r5对照组相比,细胞增殖显著增加(P<0.01);而赤芍水提液对其增殖并不显著(P>0.05)。100~500μg/ml浓度的赤芍水提物与PDGF-BB病理模型组相比,细胞增殖有显著抑制(P<0.01);而白芍水提液对其增殖并不明显(P>0.05)。结论白芍、赤芍水提物在化学成分上存在差异,并在细胞活性研究中,两者亦表现出不同的生物效应。

목적비교백작、적작수제물중소함화학성분적이동,급탐토백작、적작수제물작용우체외대서흉동맥평활기주(A7r5)적생물효응차이。방법통과HPLC급질보분석,비교백작、적작수제물중각단체화학성분적공성여차이。채용체외배양A7r5,이용혈소판원생장인자BB(PDGF-BB)유도A7r5건립세포증식병리모형,가입불동제량적백작、적작수제물,통과MTT비색법화실시세포분석의검측세포활성。결과백작、적작재화학성분상유차별,즉사량자옹유공동적성분,함량역존재차이。재A7r5증식연구현시,300μg/ml백작수제액여A7r5대조조상비,세포증식현저증가(P<0.01);이적작수제액대기증식병불현저(P>0.05)。100~500μg/ml농도적적작수제물여PDGF-BB병리모형조상비,세포증식유현저억제(P<0.01);이백작수제액대기증식병불명현(P>0.05)。결론백작、적작수제물재화학성분상존재차이,병재세포활성연구중,량자역표현출불동적생물효응。
Objective To compare the chemical components in Baishao and Chishao water extracts and investigate their effects on the proliferation of rat thoracic aorta smooth muscle cells in vitro. Methods The contents and chemical structures of monomers separated from the water extracts of Baishao and Chishao were analyzed using high-performance liquid chromatography and mass spectroscopy. Rat thoracic aorta smooth muscle cell line A7r5 and its platelet-derived growth factor-BB (PDGF-BB)-induced proliferation model were exposed to different concentrations of Baishao and Chishao water extracts, and the cell viability was analyzed by mitochondrial-dependent reduction of MTT and real-time cell analyzer. Results The growth of A7r5 cells was significantly stimulated by 300μg/ml Baishao water extract (P<0.01), but Chishao water extract produced no such effect (P>0.05). In PDGF-BB-induced cell proliferation model, the cell growth was significantly suppressed by 100-500μg/ml Chishao water extract (P<0.01), while Baishao water extract showed no obvious effect on the cell proliferation (P>0.05). Conclusion Baishao and Chishao water extracts have different chemical components and produce different biological effects.
