为了建立高效液相色谱法测定尼卡巴嗪的含量,采用Waters X-Bridge C18(150 mm ×4.6 mm,5μm)色谱柱,乙腈-水(用磷酸调pH至3.0)为流动相,梯度洗脱,流速为1.0 mL·min-1,检测波长0~5 min为297 nm,5 min开始为350 nm,柱温30℃,进样体积为20μL。2-羟基-4,6-二甲基嘧啶(HDP)的回归方程为:y=23452x-456,相关系数r=0.99999,4,4’-二硝基均二苯脲( DNC)的回归方程为:y =102388x +5296,相关系数r=0.9999,即HDP在0.29~29.12μg/mL范围内具有良好的线性, DNC在0.71~70.90μg/mL范围内具有良好的线性。该方法简便、准确、灵敏度高、重复性好,可以通过同时测定尼卡巴嗪中HDP和DNC来计算尼卡巴嗪的含量。

To develop the method of HPLC for determination of nicarbazine. The liquid chromatography separation was carried on XBridge C18 column (150 mm×4.6 mm,5μm) with acetonitrile-water(pH was adjusted to 3.0 by H3PO4) as mobile phase under the column temperature 30℃.The detection wavelength was 297 nm at 0~5 min, and 350 nm after 5 min, and the injection quantity was 20 μL . The equation of HDP was y = 23452x - 456, r=0.99999;The equation of DNC was y =102388x + 5296,r=0.9999, the linear range of HDP and DNC were 0.29~29.12 μg/mL and 0. 71~70. 90 μg/mL respectively. The method was simple , accuracy, and with high sensitivity.The content of nicarbazine can be calculated by determining HDP and DNC at the same time.
