目的:比较不同基质配方的普萘洛尔对豚鼠耳背部皮肤银屑病模型的干预作用。方法将24只豚鼠随机分为4组:空白对照组、模型基质组1、模型基质组2、模型基质组3,每组6只。空白对照组,双耳外侧每日外涂凡士林软膏,模型组用不同基质配方的5%的普萘洛尔软膏均匀涂于豚鼠双耳耳背部,每日涂3次,共20 d。10 d、20 d后分别对豚鼠耳背部皮肤进行数码相机拍照和采用游标卡尺测量耳部厚度。处死,取左耳部皮肤做病理。结果不同基质配方5%普萘洛尔干预10 d后,三组模型基质组豚鼠耳背部皮肤出现明显红斑、红点,部分出现结痂,以模型基质组2最为明显。干预20 d后,豚鼠耳背部皮肤上述现象已不十分明显。不同基质配方的5%普萘洛尔干预10 d后,与空白对照组比较,3组模型基质组豚鼠耳背部皮肤厚度明显增加,以模型基质组2增加最为明显(P<0.05)。5%普萘洛尔干预20 d后,与空白对照组比较,3组模型基质组豚鼠耳背部皮肤厚度明显增加,以模型基质组1和2增加最为明显(P<0.05)。不同基质配方的5%普萘洛尔干预20 d后,与空白对照组比较,3组模型基质组豚鼠耳背部皮肤均出现表皮增厚,明显的角化不全或角化过度,颗粒层变薄或不连续,颗粒细胞减少,棘层肥厚,真表皮交界处呈波浪状起伏。真皮层见有少量炎症细胞浸润。模型基质组2病理改变最为明显。结论不同基质配方的普萘洛尔对豚鼠耳背部皮肤均可造成银屑病模型,以5%普萘洛尔(含透皮剂)+35% PVA(成膜材料)组效果最为明显。随着干预时间的延长,病变有减轻的倾向,说明这种造模方法模拟的是银屑病急性期的病理改变。

Objective Tocomparetheinterventioneffectonpsoriasismodelofauriculardorsalskin inguineapigstreatedwithPropranoloofdifferentmatrixformulas.Methods Twentyfourguineapigswere randomized into 4 groups,named a blank control group,a model matrix group 1 (group No.1 ),a model matrix group 2(group No.2)and a model matrix group 3(group No.3),6 guinea pigs in each one.In the blank con-trol group,Vaseline ointment was used externally on the affected areas every day.In the model groups,5%Propranolo ointment of different matrix formulas were applied,3 times a day,for 20 d totally.In 10 d and 20 d,the affected areas were pictured with digital camera and the vernier caliper was used to measure the ear thickness.After sacrificed,the skin on the back of the left ear was collected for pathological observation.Re-sults In10dof5%Propranoloointmentofdifferentmatrixformulas,theredspotsandrashesappearedon the affected skin in the three model groups and scab presented in some cases,which was the most obvious in the group No.2.In the intervention of 20 d,the above symptoms were not obvious.Compared with the blank control group,in 10 d,the thickness of the affected skin was increased apparently in the three model groups, in which,the thickening was the most apparent in the group No.2 (P<0.05 ).In 20 d,compared with the blank control group,the thickness of the affected skin was increased apparently in the three model groups,in which,the thickening was the most apparent in the group No.1 and the group No.2(P<0.05).In 20 d of intervention,compared with the blank control group,the skin of the affected area was thickening,with appar-ent para-aceratosis or hyperkeratosis;the granule layer was thinning or broken,the granule cells were re-duced,acanthosis occurred,the junction of epidermis and corium was wavy.There were little amount of in-flammatory cell infiltration in corium.The most obvious pathological changes were found in the group No.2. Conclusion Propranoloofdifferentmatrixformulascaninducepsoriasismodel,andtheresultisthebest with 5% Propranolo(containing transdermal agent)+35% PVA(film -forming material).With interven-tion,the lesion is alleviated in tendency,explaining that this modeling method simulates the acute pathological changes of psoriasis.
