以实验室保藏的一株谷氨酸棒杆菌ATCC21851为出发菌株,在液体完全培养基进行活化培养,250mL三角烧瓶进行发酵培养,完成单因素试验,利用单因素试验确定了最佳的培养基pH值、装液量、接种量和培养温度。运用Box-Benhnken的中心组合设计和响应面分析,对该菌株产L-色氨酸的发酵条件组成进行多项式回归模型建立和最优化,在此基础上确定了该产 L-色氨酸菌株的最佳培养条件组成:接种量10.11CFU/mL、装液量46.25mL/250mL和培养基pH值6.76。在优化后的培养基和最佳发酵条件下进行发酵培养,测得经过发酵之后L-色氨酸产量达到9.06g/L。

Laboratory preserved corynebacterium glutamicum ATCCA21851 as starting strain, was activated in a liquid total medium, and then fermentation in 250mL erlenmeyer flask. Optimal conditions (pH value, liquid volume, in-oculum size and incubation temperature) were determined by single-factor test. Polynomial regression modeling of producing L-tryptophan strains fermentation conditions and optimization were performed by used central composite design of Box-Benhnken and response surface methodology. And optimum culture conditions were inoculum size 10.11CFU/Ml, liquid vol-ume 46.25mL/250mL and pH 6.76. L-tryptophan production was 9.06g/L in optimized medium under optimized conditions.
