目的:研究抗过敏药枸地氯雷他定对卵蛋白致敏小鼠脾淋巴细胞表达炎症因子的影响,探讨其抗过敏性炎症活性。方法小鼠腹腔注射卵蛋白联合氢氧化铝胶体构建致敏动物模型,无菌分离小鼠脾淋巴细胞,以不同浓度药物进行阻断处理,酶联免疫吸附法(ELISA)检测培养细胞上清液中白三烯(LTB4)、前列腺素E2(PGE2)、白介素4(IL-4)、白介素5(IL-5)及肿瘤坏死因子(TNF-α)的水平。结果枸地氯雷他定不同浓度可明显抑制致敏淋巴细胞释放PGE2、TNF-α、IL-4、IL-5等炎症因子,且具有浓度依赖性。结论枸地氯雷他定有显著的抗过敏性炎症活性。

목적:연구항과민약구지록뢰타정대란단백치민소서비림파세포표체염증인자적영향,탐토기항과민성염증활성。방법소서복강주사란단백연합경양화려효체구건치민동물모형,무균분리소서비림파세포,이불동농도약물진행조단처리,매련면역흡부법(ELISA)검측배양세포상청액중백삼희(LTB4)、전렬선소E2(PGE2)、백개소4(IL-4)、백개소5(IL-5)급종류배사인자(TNF-α)적수평。결과구지록뢰타정불동농도가명현억제치민림파세포석방PGE2、TNF-α、IL-4、IL-5등염증인자,차구유농도의뢰성。결론구지록뢰타정유현저적항과민성염증활성。
Objective To investigate the effect of Desloratadine Citrate Disodium on the release of several inflammatory mediators suchas leukotfiene B4 (LTB4 ),interleukin 4 (IL-4),interleukin 5 (IL-5),prostaglandin E2 (PGE2 )and tumor necrosis factor α(TNF-α)from spleen lymphocytes of allergic mice challenged by ovalbumin(OVA).Methods OVA-sensitized mice were used as animal mod-el,spleen lymphocytes isolated from mice were treated with desloratadine citrate disodium,ebastine,levocetirizine hydrochloride anddexamethasone,followed by incubation with OVA.These cells without pretreatment with drugs served as the control.Enzyme 1inked im-munosorbent assay (ELISA)was applied to measure the level of these inflammatory mediators.Results Desloratadine citrate disodiumsignificantly inhibited the release of LTB4 ,IL-4,IL-5,PGE2 and TNF-αin a dose dependent manner.Conclusion Desloratadine citratedisodium has a potent antagonistic effect on the allergic-inflammation.
