目的:观察茶多酚对创伤性颅脑损伤大鼠的脑保护作用。方法通过可控性皮层打击器建立SD大鼠颅脑损伤模型。打击24 h后对大鼠进行神经功能缺损评分,测量干湿重以及静脉注射伊文士蓝的方法评价血脑屏障(BBB,Blood-brain barri-er)的通透性,采用蛋白免疫印迹的方法测量水肿相关蛋白aquaporin-4(AQP4)和matrix metalloproteinase-9(MMP-9)的表达。结果与模型组比较,给药组打击24 h后的神经功能缺损评分明显降低(P<0.05),脑含水量和BBB开放程度明显减少(P<0.05)。给药组大鼠的水肿相关蛋白AQP4、MMP-9的表达减少(P<0.05)。结论茶多酚具有脑保护作用,其机制可能与减轻创伤后脑水肿程度,减少AQP4、MMP-9表达有关。

Objective To investigate the protective effect of tea polyphenols on traumatic brain injury.Methods The models were es-tablished by controlled cortical impact,24 hours after the strike,Neurological Severity Scores and brain water content were observed,theinjury to the blood-brain barrier was also observed by the content of Evan’s blue (EB)in the cerebral tissue,moreover,the expresstionof aquaporin-4 (AQP4)and matrix metalloproteinase-9 (MMP-9)were measured by western blot.Results Tea polyphenols improvedNeurological Severity Scores (P <0.05),alleviated brain water content and the disruption of Blood-brain barrier (BBB)(P <0.05),as well as reduced the expression of AQP4 and MMP-9 (P <0.05).Conclustion Tea Polyphenols played a role in protecting thebrain after traumatic brain injury,The potential mechanism perhaps is reducing of the extent of brain edema by inhibition of the expres-sion of AQP4 and MMP-9.
