为确定猪尿中盐酸赛庚啶的残留指示物,将15头生猪分成3组,饲养环境相同,A组为空白对照组,B组为低剂量盐酸赛庚啶组(10 mg/d),C组为高剂量盐酸赛庚啶组(100 mg/d)。对3组同时进行7 d的加量饲喂以及7d的停止饲喂体内消除实验。优化了葡萄糖醛酸酶和硫酸芳基酯酶对猪尿进行酶解反应的条件,用已经建立的UPLC-MS/MS法分别测定了酶解与非酶解的猪尿样品中盐酸赛庚啶原型药物的含量,研究猪尿中原型药物与其衍生物的积累和消除规律。实验结果表明,猪尿中盐酸赛庚啶主要以原型药物的形式存在。其含量与饲喂量呈正相关,半衰期为2 d,6 d后几乎完全消除。

To determine the residual indicator of cyproheptadine in swine urine,15 healthy swine were selected and divided into 3 groups with the same breeding environment.Group A was fed with blank feed,while group B was fed with low content(10 mg/d)and group C was fed with high content(100 mg/d).The 7 days accumulation test and 7 days elimina-tion test was held.With the glucuronidase and the sulfuricarylesterase reaction optimized as well as the UPLC-MS/MS method established,the concentration of cyproheptadine hydrochloride both enzymolyzed and unenzymolyzed was analyzed. The accumulation rule and the elimination rule were both determined.According to the result,cyproheptadine hydrochloride existed in swine urine mainly in the form of original drug.Significant correlation existed between the content and the feeding amount.The drug has a half-life of 2 days,and it almost completely eliminated after 6 days.
