目的:探讨oxHDL3对人脐静脉内皮细胞株HUVEC-12细胞组织型纤溶酶原激活物(tissue type plasminogen activator,t-PA)表达的影响。方法 HUVEC-12细胞分别经不同浓度(0、20、40、80 mg/L)HDL3、oxHDL3孵育,采用RT-PCR的方法检测t-PAmRNA表达情况。结果与0mg/L oxHDL3组比较,oxHDL3呈剂量依赖性下调t-PA mRNA的表达,以80 mg/L oxHDL3浓度组减少最明显,较0mg/LoxHDL3组下降了30%(<0.05)。结论 oxHDL3呈剂量依赖性下调人脐静脉内皮细胞t-PA mRNA的表达。

목적:탐토oxHDL3대인제정맥내피세포주HUVEC-12세포조직형섬용매원격활물(tissue type plasminogen activator,t-PA)표체적영향。방법 HUVEC-12세포분별경불동농도(0、20、40、80 mg/L)HDL3、oxHDL3부육,채용RT-PCR적방법검측t-PAmRNA표체정황。결과여0mg/L oxHDL3조비교,oxHDL3정제량의뢰성하조t-PA mRNA적표체,이80 mg/L oxHDL3농도조감소최명현,교0mg/LoxHDL3조하강료30%(<0.05)。결론 oxHDL3정제량의뢰성하조인제정맥내피세포t-PA mRNA적표체。
Objective To investigate the change of tissue type plasminogen activator mRNA expression induced by oxidized high density lipoprotein 3 .Methods HUVEC-12 were incubated with oxHDL3 at dif erent concentrations(0,10,20,40,80 mg/L)t-PA mRNA were determined by RT-PCR .Results Compared with the 0mg/L oxHDL3 group, oxHDL3 can decrease the expression of t-PA mRNA. The t-PA production decrease most significantly in the group using 80 mg/L oxHDL3 treated for 24 hours.oxHDL3 caused a significantly decreased t-PA mRNA expression by 30%, at the level of 80mg/L respectively ( <0.05). Conclusion oxHDL3 can inhibit the expression of t-PA mRNA in human umbilical vein endothelial cells.
