目的:观察IL-1β对大鼠离体幽门括约肌自发收缩的影响,初步探讨其作用机制。方法成年SD大鼠(雌雄不拘)随机分为正常组、IL-1β组、亚甲基蓝组、L-NAME组。采用离体组织灌流的方法,用RM6240 C生物信号采集系统观察IL-1β对大鼠离体幽门括约肌自发收缩作用的影响;通过应用不同信号途径的特异性阻断剂,探讨IL-1β的作用机制。结果 IL-1β(终浓度为2,20,200μg/L)能抑制大鼠离体幽门括约肌自发收缩的振幅、频率和张力。亚甲基蓝(10μmol/L)和L-NAME(100μmol/L)能部分抵消IL-1β对大鼠离体幽门括约肌自发收缩的抑制作用,与IL-1β单独作用相比,振幅、频率和张力均较高( P均<0.01)。结论 IL-1β能抑制大鼠离体幽门括约肌的自发收缩,其作用可被亚甲基蓝、L-NAME部分阻断,说明其机制很可能与cGMP、NO有关。

Objective It is to observe the effect of IL -1βon the spontaneous contractions of isolated rats ’ pyloric sphinc-ter in order to explore its mechanism .Methods Male or female SD rats were divided into 4 groups randomly:normal group , IL-1βgroup, methylene blue trihydrate group and L -NAME group, the effect of IL-1βon the spontaneous contractions of i-solated rats ’ pyloric sphincter was observed by RM 6240 C biologic signal selecting system , the mechanism of IL-1βwas ex-plored by specific repressor of different signal pathway .Results IL-1β(final concentration were 2, 20, 200μg/L) could in-hibit the amplitude , frequency and tension the spontaneous contractions of isolated rats ’ pyloric sphincter .Pretreatment of the tissues with methylene blue trihydrate (10μmol/L) and L-NAME (100μmol/L) markedly attenuated the inhibitory effects of IL-1β.The amplitude , frequency and tension were increased compared with methylene blue trihydrate absented .Pretreat-ment of the tissues with L -NAME (100μM) markedly attenuated the inhibitory effects of IL -1β.The amplitude, frequency and tension were increased compared with L -NAME absented (P<0.01).Conclusion IL-1βcan inhibit spontaneous con-tractions on rats ’ isolated pyloric sphincter .The inhibitory effects of IL -1βcan be attenuated by methylene blue trihydrate and L-NAME, which indicates that the mechanism may be related with the levels of NO and cGMP in intracellular .
