目的:探讨Bmi-1基因在人脑胶质瘤中的表达水平及其意义。方法收集2011年1月-2013年3月在郑州大学第五附属医院住院的低级别胶质瘤37例、高级别胶质瘤45例设为胶质瘤组,正常脑组织32例设为对照组。分别采用反转录-聚合酶链反应( RT-PCR)和蛋白印迹法( Western blotting)测定胶质瘤组和对照组中Bmi-1 mRNA及蛋白表达水平。结果胶质瘤组Bmi-1 mRNA及蛋白表达水平较对照组上调( P<0.05);高级别胶质瘤组Bmi-1 mRNA及蛋白表达水平较低级别胶质瘤组上调( P<0.05)。结论 Bmi-1在人脑胶质瘤中呈高表达,且与胶质瘤的病理分级相关,可能参与人脑胶质瘤的发生和恶性进展。

목적:탐토Bmi-1기인재인뇌효질류중적표체수평급기의의。방법수집2011년1월-2013년3월재정주대학제오부속의원주원적저급별효질류37례、고급별효질류45례설위효질류조,정상뇌조직32례설위대조조。분별채용반전록-취합매련반응( RT-PCR)화단백인적법( Western blotting)측정효질류조화대조조중Bmi-1 mRNA급단백표체수평。결과효질류조Bmi-1 mRNA급단백표체수평교대조조상조( P<0.05);고급별효질류조Bmi-1 mRNA급단백표체수평교저급별효질류조상조( P<0.05)。결론 Bmi-1재인뇌효질류중정고표체,차여효질류적병리분급상관,가능삼여인뇌효질류적발생화악성진전。
Objective To investigate the expression and significance of Bmi-1 in human glioma. Methods 37 cases of low grade glioma,45 cases of high grade glioma( glioma group)and 32 normal brain tissues( control group)were collected in the Fifth Affiliated Hospital of Zhengzhou University from January 2011 to March 2013. The expression of Bmi-1 mRNA and pro-tein were detected by RT-PCR and Western blotting methods. Results The expression levels of Bmi-1 mRNA and protein were significantly higher in the glioma group than in the control group(P<0. 05). The expression levels of Bmi-1 mRNA and protein were significantly higher in high-grade glioma group than in low-grade glioma group( P<0. 05 ) . Conclusion The mRNA and protein expression levels of Bmi-1 are high in human glioma and are associated with the grade of glioma. Therefore,Bmi-1 may be involved in the development and malignant progression of human glioma.
