目的:探讨糖酵解抑制剂2-脱氧-D-葡萄糖(2-DG)与氟尿嘧啶(5-Fu)单独及联合应用对人胃癌细胞MGC-803增殖及凋亡的影响。方法实验设立2-DG组、5-Fu组、联合组(2-DG+5-Fu)及空白对照组,将2-DG(10 mmol/L)及5-Fu(5.0μg/L)单药及联合用药作用于人胃癌细胞MGC-803,MTT法检测各组MGC-803细胞的增殖抑制率,流式细胞仪检测各组细胞凋亡情况。结果2-DG和5-Fu均可使胃癌细胞增殖受到抑制并可诱导细胞凋亡,两药合用后其作用明显增强。结论2-DG能有效抑制人胃癌细胞MGC-803细胞生长增殖,并诱导其凋亡;2-DG与5-Fu联合应用抗肿瘤作用明显增强。

목적:탐토당효해억제제2-탈양-D-포도당(2-DG)여불뇨밀정(5-Fu)단독급연합응용대인위암세포MGC-803증식급조망적영향。방법실험설립2-DG조、5-Fu조、연합조(2-DG+5-Fu)급공백대조조,장2-DG(10 mmol/L)급5-Fu(5.0μg/L)단약급연합용약작용우인위암세포MGC-803,MTT법검측각조MGC-803세포적증식억제솔,류식세포의검측각조세포조망정황。결과2-DG화5-Fu균가사위암세포증식수도억제병가유도세포조망,량약합용후기작용명현증강。결론2-DG능유효억제인위암세포MGC-803세포생장증식,병유도기조망;2-DG여5-Fu연합응용항종류작용명현증강。
Objective To investigate the effects of 2-deoxy-D-glucose ( 2-DG) or 5-Fu alone and the combination of both on the proliferation and apoptosis of human gastric cancer MGC-803 cells. Methods Human gastric cancer MGC-803 cells were treated with 2-DG or 5-Fu alone, or both. The inhibitory effect on the proliferation of MGC-803 cells was detected by MTT method. Apoptotic rate was assayed by flow cytometry. Results 2-DG or 5-Fu inhibited the growth of MGC-803 cells obviously and induced the apoptosis. When 2-DG and 5-Fu were combined,the inhibitory effect and apoptotic rate were improved (P < 0.05). Conclusions 2-DG inhibits the growth of human gastric cancer MGC-803 cells. The combination of 2-DG and 5-Fu improves the ability of 5-Fu to attack the tumor cells.
