【目的】优化水解醇溶蛋白生产功能肽的工艺,提高玉米醇溶蛋白的应用价值.【方法】通过正交试验获得从玉米蛋白粉中提取醇溶蛋白的最佳提取时间、提取温度、料液比、乙醇浓度等提取条件;通过碱性蛋白酶和蛋白酶K对玉米醇溶蛋白进行水解生产玉米多肽.【结果和结论】通过正交试验获得了从玉米蛋白粉中提取醇溶蛋白的最佳条件,即:料液比1∶4(g/mL),60℃水浴加热,乙醇体积分数为75%,提取3 h.两步酶解法效率高于单酶解法,两步酶解法先加蛋白酶K反应后再加碱性蛋白酶时水解效果最佳.

[Objective]The production of short peptide from zein was investigated to greatly promote the application of zein .[Method]The conditions such as extraction time , temperature , ethanol concentration and ratio of liquid to solid were optimized by orthogonal test .Zein was hydrolyzed by protease K and al-kaline protease in this study .[Result and conclusion]The optimal conditions of extracting zein were as follows:the ratio of solid to liquid was 1∶4;the extracting time was 3 hours;the extracting temperature was 60 ℃and the concentration of ethanol was 75%.Results showed that the zein could be hydrolyzed more efficiently by combining two enzymes than by only using one of the two enzymes .Zein hydrolyzed by protease K and alkaline protease in turn is the best enzymatic hydrolysis method .
