目的:研究链脲佐菌素联合烟酰胺诱导2型糖尿病大鼠模型的方法。方法 Wistar大鼠隔夜禁食,腹腔注射烟酰胺60 mg/kg,15 min后腹腔注射链脲佐菌素65 mg/kg,在诱导后72 h、2周及4周观察大鼠行为、形态学改变,测量体重,空腹检测鼠尾静脉血糖,血糖大于11.1 mmol/L的大鼠即为诱导成功。结果诱导后的大鼠出现懒动、精神萎靡症状,与空白对照组相比体重下降(P<0.05)、血糖上升(P<0.05);与单独使用链脲佐菌素诱导组相比体重上升(P<0.05)、血糖较低(P<0.05)。结论链脲佐菌素联合烟酰胺诱导糖尿病大鼠成功率高,是制备2型糖尿病大鼠模型的有效方法。
目的:研究鏈脲佐菌素聯閤煙酰胺誘導2型糖尿病大鼠模型的方法。方法 Wistar大鼠隔夜禁食,腹腔註射煙酰胺60 mg/kg,15 min後腹腔註射鏈脲佐菌素65 mg/kg,在誘導後72 h、2週及4週觀察大鼠行為、形態學改變,測量體重,空腹檢測鼠尾靜脈血糖,血糖大于11.1 mmol/L的大鼠即為誘導成功。結果誘導後的大鼠齣現懶動、精神萎靡癥狀,與空白對照組相比體重下降(P<0.05)、血糖上升(P<0.05);與單獨使用鏈脲佐菌素誘導組相比體重上升(P<0.05)、血糖較低(P<0.05)。結論鏈脲佐菌素聯閤煙酰胺誘導糖尿病大鼠成功率高,是製備2型糖尿病大鼠模型的有效方法。
목적:연구련뇨좌균소연합연선알유도2형당뇨병대서모형적방법。방법 Wistar대서격야금식,복강주사연선알60 mg/kg,15 min후복강주사련뇨좌균소65 mg/kg,재유도후72 h、2주급4주관찰대서행위、형태학개변,측량체중,공복검측서미정맥혈당,혈당대우11.1 mmol/L적대서즉위유도성공。결과유도후적대서출현라동、정신위미증상,여공백대조조상비체중하강(P<0.05)、혈당상승(P<0.05);여단독사용련뇨좌균소유도조상비체중상승(P<0.05)、혈당교저(P<0.05)。결론련뇨좌균소연합연선알유도당뇨병대서성공솔고,시제비2형당뇨병대서모형적유효방법。
Objective To explore streptozotocin -nicotinamide-induced type 2 diabetic model in rats .Methods 3-month-old Wistar rats were fasted overnight then administered 60 mg/kg nicotinamide intraperitoneally ( i.p.) fol-lowed by an i.p.injection of 65 mg/kg streptozotocin 15 min later.After induced 72 hour, 2 weeks and 4 weeks, rats behavior and body weight were observed , and the fasting blood glucose level was determined in a sample taken from the tail vein of the rats.The diabetic status was confrmed by fasting hyperglycemia (fasting blood glucose≥11.1 mmol/L). Results Ddiabetic model rats became lazy and distractive .Compared with the control group , the body weight after in-duction was lower (P<0.05) with higher blood glucose level (P<0.05);but to the group induced only by streptozoto-cin, blood glucose after induced was decreased (P<0.05) with significantly increased body weight (P<0.05).Con-clusion Streptozotocin-nicotinamide-induced diabetic rats provide an effective animal model for type 2 diabetes with high success rate .
