以硫酸软骨素的吸附率和解析率为指标,通过静态吸附实验筛选出D290阴离子交换树脂用于高效分离酶解液中的硫酸软骨素,考察了平衡时间、初始浓度、pH、温度等因素对鲨鱼硫酸软骨素分离效果的影响。较佳的分离条件如下:酶解液的初始浓度20 mg/mL,pH 5,室温,静态吸附9 h,洗脱溶剂3 mol/L NaCl溶液。在该条件下,硫酸软骨素的静态饱和吸附量(以湿树脂计)为98.9 mg/g,解析率为87.2%,硫酸软骨素产品的纯度为96.5%。D290阴离子交换树脂具有选择性好、吸附率和解析率高等优点,适用硫酸软骨素的分离纯化。

The static chondroitin sulfate adsorption and desorption ratio were used as parameters to select the resin. D290 anion-exchange resin was the best resin for separation of chondroitin sulfate from shark cartilage hydrolysates. The effects of equilibrium time, initial concentration, pH and temperature on the adsorption were investigated. The optimum separation condition was as follows:equilibrium time 9 h, initial chondroitin sulfate concentration 20.0 mg/mL, pH 5 and 25℃. The elution solvent was 3 mol/L NaCl solution. Under the condition, the static saturated adsorption capacity of chondroitin sulfate (by wet resin) was 98.9 mg/g, desorption ratio was 87.2%, and the purity of chondroitin sulfate product was 96.5%. D290 anion-exchange resin was suitable for separation of chondroitin sulfate with high adsorption and desorption ratio.
