适宜的NAD+水平,对提高以糖质原料为底物的工业发酵速度具有重要意义。以米根霉As 3.3461为研究菌株,通过改变发酵过程中溶氧量和烟酸添加量,调节胞内NADH代谢途径,胞内ATP水平随之发生改变。 ATP水平降低,糖酵解过程中关键酶PFK, HK, PK活性增加,使丙酮酸转化率增加。丙酮酸分支点关键酶LDH和ADH活性受能量代谢和糖酵解关键酶活性的影响,导致主要代谢产物L-乳酸、乙醇产量发生改变。因此,通过调节米根霉胞内NADH代谢途径,对提高L-乳酸产量具有重要意义。

Appropriate level of NAD+is important for industrial fermentation which uses sugar as substrate to improve the speed of glycolysis. By changing the amount of dissolved oxygen, and adding niacin to alter the ratios of NADH/NAD+, to study the effects made by the change of NADH metabolism for the Rhizopus oryzae As 3.3461. With the change of the NADH metabolism pathways, ATP levels change. When ATP levels reduce, the activities of PFK, HK, PK which are the key enzymes in the process of glycolysis increased, the conversion rate of pyruvate increases. Under the influence of energy metabolism and the activities of PFK, HK, PK, the yield of L-lactic acid and ethanol change. Therefore, it has important significance to improve the yield of lactic acid by regulating the NADH metabolism pathways in Rhizopus oryzae.
