目的建立以反相高效液相色谱法测定人血浆中血药浓度三唑仑的方法。方法采用Diamonsil C18(250nm×4.6nm,5μm)色谱柱,流动相为甲醇:水(60:40),流速为1.0ml/min,检测波长为234nm,柱温为25℃,采用蛋白沉淀法处理血浆样品。结果三唑仑血浆浓度在7.70~38.50μg(r=0.9997)范围内线形关系良好,低、中、高三种浓度的平均相对回收率分别为101.23%、103.88%,102.25%,RSD均小于3%。结论本方法简便、快速、准确、灵敏度高,可用于三唑仑血药浓度测定和人体药动学研究。

Objective:To establish a Rp-HPLC method for the determination of triazolam in human plasma.Method:The HPLc separation was achieved on a Diamonsil C18 column(250mm×4.6 mm,5μm),and the mobile phase consisted of MeOH-H20(60:40).The detection wavelength wan 254 nm.flow rate was 1.0ml/L.and column temperature was25℃. The protein precipitation method was used to treat the plasma samples.Result:The linear range of diazepam was7.70~38.50μg (r=0.9997),the average relative recovery of low,middle and high concentration was101.23%,103.88%and 102.25%, respectively,and the RSDs were less than 3%.Conclusion:The method is simple,rapid,accurate and sensitive for the plasma concentration monitoring and clinical pharmacokinetic studies of triazolam.
