目的研究卵清蛋白(HP-OVA)诱导阴道上皮细胞(VK2/E6E7)凋亡诱导作用及机制.方法接种VK2/E6E7细胞,分别以不同药物浓度(10、2、0.5、0mg/ml)进行处理,采用CCK8法检测细胞增殖抑制率,TUNEL细胞凋亡检测试剂盒检测细胞凋亡.结果随着HP-OVA浓度的升高, VK2/E6E7细胞增殖明显受到抑制,细胞增殖抑制率明显增加,且当HP-OVA浓度为10mg/ml时,细胞增殖抑制率达68.2%.HP-OVA诱导VK2/E6E7细胞凋亡的作用也很强,具有时间-浓度依赖性.结论卵清蛋白具有诱导阴道上皮细胞增殖抑制和促进凋亡的作用,是非常理想抗HPV的杀微生物制剂.

목적연구란청단백(HP-OVA)유도음도상피세포(VK2/E6E7)조망유도작용급궤제.방법접충VK2/E6E7세포,분별이불동약물농도(10、2、0.5、0mg/ml)진행처리,채용CCK8법검측세포증식억제솔,TUNEL세포조망검측시제합검측세포조망.결과수착HP-OVA농도적승고, VK2/E6E7세포증식명현수도억제,세포증식억제솔명현증가,차당HP-OVA농도위10mg/ml시,세포증식억제솔체68.2%.HP-OVA유도VK2/E6E7세포조망적작용야흔강,구유시간-농도의뢰성.결론란청단백구유유도음도상피세포증식억제화촉진조망적작용,시비상이상항HPV적살미생물제제.
Objective To investigate the antiviral activity of 3-hydroxyphthalic anhydride-modified ovalbumin(HP-OVA) against human papillomavirus (HPV) in vitro. Methods VK2/E6E7 cells were respectively treated with HP-OVA at different concentrations of 0,0.5,2,10 mg/ml,the anti-HPV activity against HPV virus in vitro and the cytotoxicity of HP-OVA in vaginal epithelial cells(VK2/E6E7) were detected by CCK8 colorimetric assay,and apoptotic index was determined by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL) method.Results As HP-OVA concentration increases,HP-OVA inhibited VK2/E6E7 cell growth significantly. We found that when the final concentration of EDF was 10mg/ml,the cell growth inhibitory rate of the cells was 68.2%.HP-OVA also can induce VK2/E6E7 cell apoptosis in in dose dependent and time dependent manners. Conclusion HP-OVA has the proliferation inhibition and apoptosis induction ,which was an excellent microbicides against HPV infection.
