目的研究褪黑素在 Beagle 犬体内药代动力学参数.方法选6只Beagle 犬行单剂口服褪黑素(3mg/只),高效液相色谱法测定血中药物浓度,经3p97程序推算药动学参数.结果褪黑素在体内药-时曲线为二室模型.主要药动学参数t1/2Ka 为(0.16±0.10) h, t1/2Ke 为(1.21±0.52) h, tmax为(0.50±0.18) h, C max 为(11.27±3.77) ng / ml, AUC(0→T)为(25.23±7.71)(ng?h)/ ml.结论褪黑素在犬体内口服吸收快、作用迅速,但维持时间较短.

OBJECTIVE: To study pharmacokinetic parameters of melatonin tablet in beagle dogs. METHODS:A single oral dose of 3mg melatonin tablet was given to 6 beagle dogs, drug concentrations in plasma were determined by HPLC method, the pharmacokinetic parameters were calculated by 3p97 pharmacokinetic program. RESULTS: The concentration-time curves of melatonin fitted two compartment model in the beagle dogs. The main pharmacokinetic parameters t 1 / 2K a、t 1 / 2K e、t max、C maxand AUC ( 0 ~ T ) were (0.16±0.10) h、(1.21±0.52) h、(0.50±0.18) h、(11.27±3.77) ng / ml and(25.23±7.71) (ng?h) / ml respectively. CONCLUSION: It is rapid to absorb melatonin and initiate sleep in beagle dogs, but it 's hold- time is short.
