目的:研究尿路致病性大肠埃希菌Ⅰ型菌毛重组质粒pcDNA3.0-fimH产生的免疫反应及其对小鼠的免疫保护作用。方法选取BALB/c小鼠60只,随机分为3组( PBS对照组、空质粒对照组、疫苗免疫组),3组分别用构建成功的重组质粒pcDNA3.0-fimH(为疫苗免疫组),载体质粒(pcDNA3.0)(为空质粒对照组)和PBS液(为PBS对照组),经股四头肌注射免疫小鼠,于第21天和第35天加强免疫。于免疫第0、14、28及42天收集小鼠膀胱冲洗液,检测各组SIgA水平。于免疫第42天采血,检测各组特异性IgG、IgG1、IgG2a水平。结果免疫后,疫苗免疫组小鼠膀胱灌洗液中sIgA水平随时间延长呈上升趋势,且明显高于PBS对照组及空质粒组(F=10.08,P<0.05);疫苗免疫组小鼠血清特异性IgG、IgG1、IgG2a抗体均明显高于对照组及空质粒组(P<0.05);结论 pcDNA3.0-fimH基因疫苗可诱导BALB/c小鼠产生特异性体液免疫,对小鼠尿道具有一定的免疫保护作用。

목적:연구뇨로치병성대장애희균Ⅰ형균모중조질립pcDNA3.0-fimH산생적면역반응급기대소서적면역보호작용。방법선취BALB/c소서60지,수궤분위3조( PBS대조조、공질립대조조、역묘면역조),3조분별용구건성공적중조질립pcDNA3.0-fimH(위역묘면역조),재체질립(pcDNA3.0)(위공질립대조조)화PBS액(위PBS대조조),경고사두기주사면역소서,우제21천화제35천가강면역。우면역제0、14、28급42천수집소서방광충세액,검측각조SIgA수평。우면역제42천채혈,검측각조특이성IgG、IgG1、IgG2a수평。결과면역후,역묘면역조소서방광관세액중sIgA수평수시간연장정상승추세,차명현고우PBS대조조급공질립조(F=10.08,P<0.05);역묘면역조소서혈청특이성IgG、IgG1、IgG2a항체균명현고우대조조급공질립조(P<0.05);결론 pcDNA3.0-fimH기인역묘가유도BALB/c소서산생특이성체액면역,대소서뇨도구유일정적면역보호작용。
Objective To investigate the immunoreactions of the recombinant plasmid pcDNA 3.0-fimH of type 1 pilus of uropatho-genic Escherichia coli and its immune protection to mice .Methods A total of 60 BALB/c mice were selected and divided into 3 groups randomly(a PBS group, a pcDNA3.0 group, an experimental group.Mice in the three groups were inttamusculary injected 100μl of recombinant plasmid pcDNA 3.0-fimH or pcDNA3.0 or PBS.The immunity of mice was strengthened at the twenty-first day and the thirty-fifth day.On days 0,14,28 and 42 d after first vaccination,bladder lavage fluid was collected to detect SIgA level in each group.And blood were sampled on 42d after first vaccination to detect IgG , IgG1, IgG2a level in each group.Results After the immu-nization,the level of specific sIgA antibody in the bladder lavage fluid of experimental group presented a tendency of increasing .And the level of specific sIgA antibody are higher than that of comparison teams (F=10.08,P<0.05).The level of specific IgG, IgG1, IgG2a antibody in the blood-serum of the mice of the experimental group is higher than control group and pcDNA 3.0 group ( P<0.05).Conclusion Recombinant pcDNA3.0-fimH vaccine can lead BALB/c mice to produce specific humoral response and have an immune protection to the mice'urethra.
