Lung%ischemia-perfuse injury%Fang Ji Huang Qi Tang(防己黄芪汤)%pathological examination
目的:研究防己黄芪汤对兔肺缺血再灌注损伤的保护。方法:36只日本大耳白兔,随机分为3组,每组12只。A组为模型对照组:建立兔肺缺血再灌注模型(左肺缺血1 h,再灌注4~6 h)。B组为黄芪汤治疗组:自模型建立前一周开始,每日喂饮中药黄芪汤2 mL/kg。C组为防己黄芪汤治疗组:自模型建立前一周开始,每日喂饮中药防己黄芪汤2 mL/kg。再灌注后4 h及6 h,分别检测肺湿/干重比(wet to dry weight ratio,W/D)、肺损伤定量指数(the index of quantitative assessment of lung histological injury, IQA)、肺光镜病理、肺电镜超微病理、肺caspase-3含量免疫组化测定。结果:再灌注后4 h(T4)及6 h(T6), B组、C组的W/D及IQA值,同A组比较,明显地下降,其中以C组下降更明显(B组:P<0.05, C组:P<0.01)。B、C两组肺组织病理学结构破坏较A组明显改善,尤以C组改变明显。肺组织电镜观察,B、C两组较A组破坏较小,肺泡结构相对完整。B、C两组caspase-3表达较A组明显降低,P<0.05。结论:防己黄芪汤能明显减轻兔肺缺血再灌注损伤,对肺组织有保护作用。

목적:연구방기황기탕대토폐결혈재관주손상적보호。방법:36지일본대이백토,수궤분위3조,매조12지。A조위모형대조조:건립토폐결혈재관주모형(좌폐결혈1 h,재관주4~6 h)。B조위황기탕치료조:자모형건립전일주개시,매일위음중약황기탕2 mL/kg。C조위방기황기탕치료조:자모형건립전일주개시,매일위음중약방기황기탕2 mL/kg。재관주후4 h급6 h,분별검측폐습/간중비(wet to dry weight ratio,W/D)、폐손상정량지수(the index of quantitative assessment of lung histological injury, IQA)、폐광경병리、폐전경초미병리、폐caspase-3함량면역조화측정。결과:재관주후4 h(T4)급6 h(T6), B조、C조적W/D급IQA치,동A조비교,명현지하강,기중이C조하강경명현(B조:P<0.05, C조:P<0.01)。B、C량조폐조직병이학결구파배교A조명현개선,우이C조개변명현。폐조직전경관찰,B、C량조교A조파배교소,폐포결구상대완정。B、C량조caspase-3표체교A조명현강저,P<0.05。결론:방기황기탕능명현감경토폐결혈재관주손상,대폐조직유보호작용。
Objective To study the protective effects of Fangji-Huangqi-Tang(防己黄芪汤) on the lung ischemia-reperfusion injury in rabbits. Methods A total of 36 Japanese long-eared rabbits were randomly di-vided into three groups (12 in each). In group A,rabbits underwent the left lung ischemia-reperfusion. The lungs were perfused for 4~6 h after 1h ischemia. In group B, rabbits were treated with Huangqi-Tang(黄芪汤) 2 mL· kg-1 · d-1 for a week before they underwent the injury model. In group C, rabbits were treated with Fangji-Huangqi-Tang(防己黄芪汤) 2 mL·kg-1·d-1 for a week before they underwent the injury model. Then we examined the wet to dry ratio in lung’s weights (W/D), the index of lung histological injury (IQA), pathology changes of lungs under the light microscope and ultramicro-pathological changes of lungs under the electron mi-croscope. At the same time, we also examined lungs’Caspase-3 expression by immunohistochemistry detection. Results At the 4th (T4) and 6th hours (T6) after reperfusion, W/D and IQA of the group B and C were obviously decreased while compared with the A group, P<0.05. On pathological changes at the 4th and the 6th hours af-ter reperfusion, compared with the group A, group B and group C had more improvements on the disorganization of lungs and on the pulmonary alveoli’s formation. On the immunohistochemistry, the levels of the Caspase-3 ex-pression in the group B and the group C were less than that of group A. On all above measurements, the chang-es in group C were more obvious compared with group B. Conclusion Fangji-Huangqi-Tang(防己黄芪汤) could reduce the ischemia-perfuse injury in rabbits’lungs. It can provide a good protection on the organization and the ultra-structure of rabbit lungs.
