目的观察比较正常氧和低氧环境下,不同细胞生长因子对犬骨髓间充质干细胞迁移的影响。<br> 方法抽取犬骨髓,采用密度梯度离心法分离骨髓间充质干细胞并进行培养。在倒置显微镜下观察细胞的形态变化;取第3代细胞利用流式细胞术检测其表型特征;加入碱性成纤维细胞生长因子、血管内皮生长因子和胰岛素样生长因子-1,在正常氧和低氧条件下行迁移和划痕实验来观察细胞趋化能力。<br> 结果细胞迁移情况与氧环境及细胞生长因子的存在和种类均有关联。碱性成纤维细胞生长因子、血管内皮生长因子和胰岛素样生长因子-1均能促进骨髓间充质干细胞的迁移,碱性成纤维细胞生长因子较其他细胞生长因子显示出更强的促进迁移作用,这种作用在低氧环境下更为明显。<br> 结论低氧环境下骨髓间充质干细胞的迁移能力较正常氧环境下更强,碱性成纤维细胞生长因子、血管内皮生长因子和胰岛素样生长因子-1的存在均有利于骨髓间充质干细胞的迁移。
目的觀察比較正常氧和低氧環境下,不同細胞生長因子對犬骨髓間充質榦細胞遷移的影響。<br> 方法抽取犬骨髓,採用密度梯度離心法分離骨髓間充質榦細胞併進行培養。在倒置顯微鏡下觀察細胞的形態變化;取第3代細胞利用流式細胞術檢測其錶型特徵;加入堿性成纖維細胞生長因子、血管內皮生長因子和胰島素樣生長因子-1,在正常氧和低氧條件下行遷移和劃痕實驗來觀察細胞趨化能力。<br> 結果細胞遷移情況與氧環境及細胞生長因子的存在和種類均有關聯。堿性成纖維細胞生長因子、血管內皮生長因子和胰島素樣生長因子-1均能促進骨髓間充質榦細胞的遷移,堿性成纖維細胞生長因子較其他細胞生長因子顯示齣更彊的促進遷移作用,這種作用在低氧環境下更為明顯。<br> 結論低氧環境下骨髓間充質榦細胞的遷移能力較正常氧環境下更彊,堿性成纖維細胞生長因子、血管內皮生長因子和胰島素樣生長因子-1的存在均有利于骨髓間充質榦細胞的遷移。

Objective This study was aimed to observe and compare the influences of cell growth factors on the migration of bone mesenchymal stem cells (BMSCs) of canines under normoxia and hypoxia. <br> Methods BMSCs were obtained from canines’ bone marrow using density gradient centrifugation. Their growth status was observed under the inverted microscope continuously. Their phenotypic characteristics of the third generation of BMSCs were analysed through flow cytometry, and their migration ability under the normal and hypoxia environment with basic fibroblast growth factor, vascular endothelial growth factor and insulin-like growth factor-1 was measured through Boyden chamber methods and wound healing assay. <br> Results There was relevance between the cell migration and oxygen content, the presence or absence of cell growth factors and their kinds. Basic fibroblast growth factor, vascular endothelial growth factor and insulin-like growth factor-1 can promote BMSCs to migration. Among them, basic fibroblast growth factor showed stonger promotion of the migration of BMSCs than others, especially under hypoxia environment. <br> Conclusion The ability of migration of BMSCs is better under hypoxia environment compaired with normal oxygen environment. The existence of basic fibroblast growth factor, vascular endothelial growth factor and insulin-like growth factor-1 is beneficial to the migration of BMSCs, which can provide reference of the research of micro environmental conditions of regenerative medicine.
