目的:观察子宫肌瘤与正常子宫肌层中成纤维细胞向成肌纤维细胞转分化的差异。原代培养人子宫肌瘤成肌纤维及鉴定。方法:(1)收集人子宫肌瘤、正常子宫肌层组织标本各30例;(2)采用免疫荧光三标鉴别正常子宫肌层与肌瘤组织中成肌纤维细胞的差异;(3)对人子宫肌瘤组织中的成肌纤维细胞进行原代分离培养,免疫细胞化学进行鉴定。结果:免疫荧光证实子宫肌瘤组织中成肌纤维细胞数较正常子宫肌层多,具有统计学意义(P<0.05)。子宫肌瘤组织成肌纤维细胞原代分离培养成功,免疫细胞化学结果显示所培养的细胞呈 Vimentin、α-SMA阳性表达,H-Caldesmon阴性表达,证实为成肌纤维细胞。结论:子宫肌瘤中成纤维细胞向成肌纤维细胞的转化较正常子宫肌层明显活跃,可能是子宫肌瘤发展过程中的关键因素之一。

목적:관찰자궁기류여정상자궁기층중성섬유세포향성기섬유세포전분화적차이。원대배양인자궁기류성기섬유급감정。방법:(1)수집인자궁기류、정상자궁기층조직표본각30례;(2)채용면역형광삼표감별정상자궁기층여기류조직중성기섬유세포적차이;(3)대인자궁기류조직중적성기섬유세포진행원대분리배양,면역세포화학진행감정。결과:면역형광증실자궁기류조직중성기섬유세포수교정상자궁기층다,구유통계학의의(P<0.05)。자궁기류조직성기섬유세포원대분리배양성공,면역세포화학결과현시소배양적세포정 Vimentin、α-SMA양성표체,H-Caldesmon음성표체,증실위성기섬유세포。결론:자궁기류중성섬유세포향성기섬유세포적전화교정상자궁기층명현활약,가능시자궁기류발전과정중적관건인소지일。
Objective :To observe the difference of the fibroblasts transdifferentiate into myofibroblasts between uterine fibroids and normal myometrium ,culture and identification of the primary cells of uterine fibroids myofibroblast .Methods (1 ) A total of 30 human uterine fibroids samples and another 30 normal myometrium were collected .(2) The difference of myofibroblasts between normal myometrium and leiomyoma were distinguished by immunofluorescence ;(3) Myofibroblast in human leiomyoma was established by primary culture ,and was tested by immunocytochemical method .Results The myofibroblasts in leiomyoma tissue more than in normal myometrium (P < 0 .05 ) . The primary leiomyoma myofibroblasts was isolated and cultured successfully , and proved by the immunocytochemistry results showed that in primary cells the expression of Vimentin and α-SMA positive ,meanwhile the expression of H -Caldesmon negative . Conclusion The uterine fibroids fibroblasts transdifferentiate into myofibroblasts than in normal myometrium ,which may be one of the key factors in the development process of the uterine fibroids .
