目的探讨左旋—二脱水伊地醇双甲磺酸酯(DMDAI-L)对HL-60细胞Bax蛋白表达的影响。方法通过免疫细胞化学法和western blot分别测定不同剂量DMDAI-L对HL-60细胞的Bax蛋白的表达情况。结果免疫细胞化学法定性实验结果显示药物干预后HL-60细胞Bax阳性表达高于正常对照组;western blot结果显示不同浓度药物(2.5μg/ml、5μg/ml、10μg/ml)干预HL-60细胞48h后,Bax蛋白相对表达(0.59±0.02、0.90±0.09、1.07±0.03),正常对照组为(0.32±0.03);各给药组与正常对照组比较均有统计学意义(p<0.05)。结论 DMDAI-L诱导HL-60细胞凋亡过程中,其作用机制可能与上调Bax蛋白表达有关。

Objective The aim of this present research was to investigate the expression of the apoptosis-related protein Bax in HL-60 cells after treatment with DMDAI-L.Methods HL-60 was treated with different doses of DMDAI-L for 48 hours. The protein of Bax was determined by immunocytochemical method and western blot analysis.Results The positive expression of Bax was higher than control group after treatment with DMDAI-L about 48 hours;For western blot ,the ratio of Bax protein were (0.59±0.02、 0.90±0.09 、1.07±0.03) and the control one was (0.32±0.03). There was significant difference between control group and experience groups (p<0.05).Conclusion The ability of DMDAI-L induced HL-60 cells apoptosis may be related to increase Bax protein expression level.
