目的::研究自发性高血压大鼠(SHR)心肌细胞凋亡与Bcl-1/Bax蛋白表达的关系,探讨星状神经节阻滞对SHR心肌细胞凋亡的保护作用。方法:将32只10周龄雄性SHR随机分为4组(n=8):左侧星状神经节阻滞组(LS组)、右侧星状节阻滞组(RS组)、药物卡托普利组(D组)、手术对照组(C组)。用ALC-NIBP无创血压测量系统测定大鼠血压,第10周实验结束后用3%戊巴比妥钠腹腔注射(45 mg/kg)麻醉SHR,取出心脏用TUNEL法测定左室心肌细胞凋亡指数,免疫组织化学检测心肌Bcl-2和Bax表达的含量。结果:与C组和LS组比较,RS组SHR凋亡指数明显降低(P<0.05);心肌Bcl-2表达明显增强(P<0.05),Bax表达明显减弱(P<0.05),Bcl-2/Bax比值增高(P<0.05)。结论:Bcl-2抗凋亡及Bax促凋亡蛋白表达参与SHR心肌细胞凋亡;右侧星状神经节阻滞能够影响凋亡相关基因蛋白的表达,降低心肌细胞凋亡指数,减轻左心室肥厚。
目的::研究自髮性高血壓大鼠(SHR)心肌細胞凋亡與Bcl-1/Bax蛋白錶達的關繫,探討星狀神經節阻滯對SHR心肌細胞凋亡的保護作用。方法:將32隻10週齡雄性SHR隨機分為4組(n=8):左側星狀神經節阻滯組(LS組)、右側星狀節阻滯組(RS組)、藥物卡託普利組(D組)、手術對照組(C組)。用ALC-NIBP無創血壓測量繫統測定大鼠血壓,第10週實驗結束後用3%戊巴比妥鈉腹腔註射(45 mg/kg)痳醉SHR,取齣心髒用TUNEL法測定左室心肌細胞凋亡指數,免疫組織化學檢測心肌Bcl-2和Bax錶達的含量。結果:與C組和LS組比較,RS組SHR凋亡指數明顯降低(P<0.05);心肌Bcl-2錶達明顯增彊(P<0.05),Bax錶達明顯減弱(P<0.05),Bcl-2/Bax比值增高(P<0.05)。結論:Bcl-2抗凋亡及Bax促凋亡蛋白錶達參與SHR心肌細胞凋亡;右側星狀神經節阻滯能夠影響凋亡相關基因蛋白的錶達,降低心肌細胞凋亡指數,減輕左心室肥厚。
목적::연구자발성고혈압대서(SHR)심기세포조망여Bcl-1/Bax단백표체적관계,탐토성상신경절조체대SHR심기세포조망적보호작용。방법:장32지10주령웅성SHR수궤분위4조(n=8):좌측성상신경절조체조(LS조)、우측성상절조체조(RS조)、약물잡탁보리조(D조)、수술대조조(C조)。용ALC-NIBP무창혈압측량계통측정대서혈압,제10주실험결속후용3%무파비타납복강주사(45 mg/kg)마취SHR,취출심장용TUNEL법측정좌실심기세포조망지수,면역조직화학검측심기Bcl-2화Bax표체적함량。결과:여C조화LS조비교,RS조SHR조망지수명현강저(P<0.05);심기Bcl-2표체명현증강(P<0.05),Bax표체명현감약(P<0.05),Bcl-2/Bax비치증고(P<0.05)。결론:Bcl-2항조망급Bax촉조망단백표체삼여SHR심기세포조망;우측성상신경절조체능구영향조망상관기인단백적표체,강저심기세포조망지수,감경좌심실비후。
Objective:To investigate the relationship between apoptosis of myocardial cells in spontaneously hypertensive rats (SHRs) and the expressions of Bcl-2 and Bax protein, and the protective effect of stellate ganglion block on apoptosis of myocardial cells.
<br> Methods:A total of 32 ten-week-old male SHRs were assigned randomly into 4 groups:a left stellate ganglion block group (group LS), a right stellate ganglion block group (group RS), a captopril group (group D) and a control group (group C). The arterial systolic blood pressure was measured by ALC-NIBP system. After 10 weeks, all rats were anaesthetized by 3%pentobarbital sodium, cardiomyocyte apoptosis index of left ventricle was assessed by TUNEL, and the localization of myocardium Bcl-2, Bax was investigated by immunohistochemistry.
<br> Results:Compared with group LS and C, the apoptotic index decreased (P<0.05). SHR myocardial expression of Bcl-2 signiifcantly increased (P<0.05), Bax expression signiifcantly decreased (P<0.05) and Bcl-2/Bax was signiifcantly higher (P<0. 05) in group RS.
<br> Conclusion:Bcl-2 and Bax play an important role in the apoptosis of myocardial cells in SHRs. Right stellate ganglion block can reduce the apoptosis of myocardial cells and reverse the reconstruction of the left ventricle in SHRs via regulation of apoptosis-related gene proteins.
