为了把测试分析误差控制在允许范围内,保证测试结果有一定的精密度和准确度,使分析数据在给定的置信水平内达到要求的质量,根据JJF 1059─1999《测量不确定度评定与表示》技术规范,估算了转基因水稻外源成分NOS终止子含量测定结果的不确定度。 A类评估结果为uA =0.001,B类评估结果为uB =0.008,合成不确定度为uC =0.008,置信水平为95%的扩展不确定度为U95=0.016,NOS终止子含量测量结果为0.1124±0.016。本实验测量结果不确定度分析表明,转基因水稻外源基因NOS终止子含量测定的不确定度主要来自于实验操作中的随机效应、微量移液器转移液体的误差、Ct值与lgA值的线性偏离。因此,在转基因生物安全定量分析中应重点注意上述不确定度来源对测量结果的影响。

In order to control the error of test analysis within the scope of the permit , to ensure that the test results are of cer-tain accuracy and precision, and to make that the analysis data achieve the required quality within a given confidence level , accord-ing to the technical specifications of “evaluation and expression of measurement uncertainty” (JJF 1059─1999), we estimated the uncertainty of content detection result of NOS terminator from foreign gene in genetically modified rice .The calculation results of measurement uncertainty showed that the type A uncertainty (uA ) was 0.001, type B uncertainty (uB ) was 0.008, compound un-certainty (uC ) was 0.008, expanded uncertainty (U95 ) at 95% confidence level was 0.016, and the measurement result of NOS terminator was 0.1124 ±0.016.It could be concluded that the measurement uncertainty of NOS terminator from foreign gene in ge-netically modified rice was mainly derived from the random effect of experimental operation , the error of transferring liquid by using micropipette, and the linear deviation of Ct value versus lgA value.Therefore, in the quantitative analysis of the safety of genetically modified organisms, the more attention should be paid to the sources of uncertainty mentioned above .
